Collagen is the most important component of the extracellular matrix,and it is a non branched fiber protein.Collagen is white,opaque,which composed of three a chains coiling helical structure,and its molecular weight is about 300KDa.Effects of collagen in animal cells is a combination of body tissues.There is a large number of collagen in aquatic invertebrates,whose type is I and V,and it have the same effect with collagen I in vertebrates.Chitosan is a product of acetylation reaction of chitin,which is a kind of shiny and translucent solid sheet.Chitosan is insoluble in water or alkali.but it has much advantages,such as.good biological compatibility,safety and biodegradability.Prepare the collagen/chitosan composite,and make research of its physical and chemical properties,the effect on melanin synthesis and antioxidant activity out of body is also include.Method:1.Use Ionic gelation method to prepare collagen/chitosan composite;use scanning electron microscope to watch the morphology of composite;use DSC,XRD.FTIR to detect the physicalstructure of composite;make research of the water retention,wettability and oil absorption of composite.2.Use kit to detect the activity of SOD and the content of MDA to explore the impact of composite on antioxidant activity of mice.3.Use murine B16 melanoma cells as model to study the effects on melanoma cells of different concentration of collagen and composite;use microplate to detect the content of melain;detect the activity of Tyrosine enzyme according to foreign literature;use RT-PCR to observe the mRNA expression of Tyrosine enzyme.Result:1.We successfully prepared collagen/chitosan composite,and the data from DSC,DSC.XRD,FTIR could support this result.2.Experiment of mice showed that composite do well in antioxidant activity,which decrease the content of MDA and increase the activity of SOD.3.According to the experiment on mice B16 melanoma,composite we have made could effectively reduce the content of melain in cells,decrease the activity of Tyrosin enzyme and make less the mRNA expression of Tyrosin enzyme.Conclusion:This topic use Ionic gelation method successfully prepared collagen/chitosan composite.According to a series of studies that show that composite do well in antioxidant activity.At the same time,composite could effectively reduce the content of melain in cells,decrease the activity of Tyrosin enzyme and make less the mRNA expression of Tyrosin enzyme.
