Panax notoginseng is a traditional common medicinal herb of our country,containing a variety of effective components and having extensive physiological functions.It's a common oral Chinese medicine,which has a special bitterness,poor taste.The main effective components of total saponinsits are easy to decompose,because of its poor light and thermal stability.Quercetin,as its main flavonoids,poorly solubles in water,so it is difficult for the body to absorb.Currently,market leading products of notoginseng which continue to use single herb preparation that processed by mechanical crushing,have exact curative effects,but the amount is larger.Superfine grinding technology has beginned to apply in notoginseng preparations,but there is still no consistent conclusions on it.There are no reports about ?-cyclodextrin used in traditional Chinese medicine(TCM)herb preparation all there,though it has been widely used in the field of TCM.This article uses the cyclodextrin inclusion technique and superfine grinding technology to develop notoginseng preparation of products,in order to provide a good water solubility,stability and pharmaceutical performance good preparation products.We study molecular properties of notoginseng total saponin before and after purification through spectral methods.We found that molecular associations of total saponins before purification are obvious in the solution,and effects of ?-cyclodextrin on the molecular association are studied.The results showed that the ?-cyclodextrin can eliminate this molecular association.Besides,computer aided technology was use to build the structure of notoginseng saponin R1 and molecular associations.Further study found that the inclusion formation of ?-cyclodextrin with total saponins is strong in the solution,and apparent binding constant is 464 L·mol-1.This shows that stable inclusion complex was formed between ?-cyclodextrin and total saponins,which is the important reason for the elimination of total saponins of associating.In addition,?-cyclodextrins with flavone and proteins also has good apparent inclusion constant.?-cyclodextrin-notoginseng powder are prepared successfully based on the inclusion associations of cyclodextrins and mechanical crushing,and then Transmission electron microscopy(TEM)was utilized to analyze it.Besides,TA and IR were applied to identify the formation of inclusion complexes in ?-cyclodextrin-notoginseng micro powders.Inspecting micronization effects through solubilization and influencing factors of experiment examining.The results show that the ?-cyclodextrin-notoginseng micro powders can improve properties of the powder pharmacy.Granules and tablets produced with these micro powders are all conformed to the requirements of pharmacopoeia,and they are better than some existing formulations in the stability of total saponins?dissolution rate and appearance.This study successfully developed a new method of improving notoginseng drug product to solve the shortage of the existing technology by combining the supramolecular inclusion technology with micronization technology.A technology with simple operation process,good product performance and lower cost is provided for industrial production,also provide a new method and train of thought to better development and utilization of panax notoginseng.
