1 ObjectiveThis project takes Zuojin Pill as the object,establishes HPLC fingerprint of Zuojin Pill and similar recipes,and establishes hot-syndrome of gastric mucosal lesion rat models by filling the stomach method.The project collectes Characteristic peaks and pharmacodynamics data of Zuojin Pill and similar recipes,analyzes the variable matrix through the partial least squares(PLS)regressionmethod to construct the fictional model between the corresponding X,Y and explores the spectrum-effect relationship of the chemical components represented by characteristic common peak and pharmacodynamic index of the recipes.2 Methods2.1 Establishes HPLC fingerprint of Zuojin Pill and its similar recipesTo establish HPLC fingerprint of Zuojin Pill and similar recipes,through optimizing chromatographic conditions,and making systematic methodology investigation.To research the total level correlation of the fingerprint by using the similarity degrees between the HPLC fingerprint as the evaluation index.2.2 Investigation of fingerprint based on chemical metrological law and recognition of dissolution pattern rulesConstruct variable matrix with the relative dissolution rate of Zuojin Pill and similar recipes,and extract the principal component factor through principal component analysis method.Conduct systematic cluster analysis choosing the principal component factor as a variable matrix and construct new variable matrix.Establish six Fisher discrima models,by using result of the systematic analysis as discriminant basis to progress Fisher discriminant analysis.2.3 The research of the evaluation of pesticide effect on Zuojin Pill and its similar prescription on hot-syndrome of gastric mucosal lesion rats modelsEstablish hot-syndrome of gastric mucosal lesion rats models,using impairment index of gastric mucosal and infiltration of inflammatory cell as indicator to evaluate the pesticide effect.2.4 The associated research of HPLC fingerprint of Zuojin Pill and its similar prescription and pharmacodynamicMake variable matrix by using characteristicpeaks(X)of HPLC fingerprint of Zuojin Pill and its similar prescription and data of pharmacodynamics(Y).To study the direction of action and contribution of the each component of similar prescription,making the PLS analys of the variable matrix by SIMCA-P 11.5 software,and investigating the relation between the characteristic peaks and pharmacodynamics.3 ResultsThe study establishes the HPLC fingerprint of Zuojinwan and its similar formulaes preliminarily.Divide three principal components through principal component analysis.Basing on the principal component analysis,divide the dissolved rule of the components of Zuojinwan and its similar formulaes into six classes through cluster analysis,and establish the corresponding Fisher discriminant function,the return sentence accuracy rate of which is 100%;Experiment results show that the Zuojin Pill is the best obviously effective therapy to hot-syndrome of gastric mucosal lesion rats models;Establish PLS model,using the parameters of the w1*c1/w2*c2 plane,CoeffCS and VIP to value.There is high-positivecorrelation between two pharmacodynamics indicator with characteristic peaks number 10,12,9,18,19,15,17,14 in hot-syndrome of gastric mucosal lesion rats models,in order of height of correlation.4 ConclusionDifferent material compatibility proportion can lead to the significant difference between the material foundation and pharmacological effects and efficacy of Zuojinwan and its similar formulaes.This study also tests and verifies the the principle of treatment according to differentiation of symptoms and signs in TCM theory,and explore and lay the foundation for the association of the change of scale of compatibility with pharmacodynamic to hot-syndrome of gastric mucusal lesion rats models,and offer analytical methods and theoretical basis for the direction of the development of the spectrum-efficient research of Zuojinwan from the vague,general model to quantitative and visualized model.
