Objective: In ordor to reform the form of prepared druds for formula of Zuojin Pill,optimizing the extraction technology for it. Methods: 1. With the total extractingsubstances,rutaecarpine as the indexes, optimizing the supercritical fluid extraction(SFE-CO2) technologic condition for Fructus evodiae by the method of orthogonal design andcomparing with vapour distillation of Fructus evodiae. 2. With the berberine,totalalkaloids and dry extract as the colligation indexes: (1) optimizing the semi-bionicextraction(SBE) technologic condition for formula of zuojin pill by the method ofuniform-design. (2) optimizing alcohol precipitation concentration of SBE solution forformula of Zuojin Pill by the method of proportion division. (3)comparing the colligationindexes Y of five kinds extracting solution for formula of Zuojin Pill[semi-bionicextraction(SBE), water extraction(WE),. alcohol extraction (AE), semi-bionic extractionand then alcohol precipitation(SBAE), water extraction and then alcoholprecipitation(WAE)]. 3.comparing the Pharmacodynamics of anti-bacteria, anti-inflammation on SBE. AE, WE. Results: 1.In the SFE- CO2 technologic condition forFructus evodiae, influencing sequence of each factor(A pressure. B temperature,C carryagent,D time) on effective ingredient of Fructus evodiae is C>A>D>B, and C(carry agent)is the remarkable ingredient (P 0.05). 2. The SFE-CO2 is better than vapour distillationfor extracting effective ingredient of Fructus evodiae.3. The technologic condition ofSBE was: PH in first extractiong was adjusted to 2.0 by the use of HCL soluteion. Andthen PH in second and third extraction was adjusted to 7.0 and 9.0 by the use of NaOHsoluteion respectively, Duration of run was 170min.85min.and 40 min.respectivel. 4. Theoptimizing alcohol precipitation concentration of SBE solution for formula of Zuojin Pill Is 55% .5. The sequence of the colligation indexes Y is SBE>WE>AE>SBAE>WAE. SBE isthe better technologic condition for formula of Zuojin Pill. 6.The effects of anti-bacteriaon Zuojin Pill extracting solution are SBE>WE>AE; The effect of anti-bacteria togolden-yellow grape coccous is better than to large intestine bacilus. 7. Zuojin Pillextracting solution have significant inhibitory effects on mice ear swelling induced byxylene. The sequence of the inhibitory ratio is SBE>WE>AE. Conclusion: whenreforming the form of prepared druds for formula of Zuojin Pill,the better technologiccondition for it are:Fructus evodiae, the first, was extracted by SFE- CO2, and the residuewas mixed with Rhizoma Coptidis and then extracted by SBE. The technologic condition ofFructus evodiae by SFE- CO2 was: pressure was 26mpa, temperature was 40℃, carry agentwas 95%alcohol, extracting time was 1.5h. The technologic condition of SBE was: PH infirst extractiong was adjusted to 2.0 by the use of HCL soluteion. And then PH in secondand third extraction was adjusted to 7.0 and 9.0 by the use of NaOH soluteionrespectively. Duration of run was 170min,85min,and 40 min,respectivel. The effects ofanti-bacteria,anti-inflammation on Zuojin Pill also show that the better technologiccondition for formula of Zuojin Pill are:Fructus evodiae, the first, was extracted by SFE-CO2, and the residue was mixed with Rhizoma Coptidis and then extracted by SBE.
