With the wide applications of fluorescent probes in different fields of life,the research on fluorescent probes gradually deepen.Nitro reductase overexpressing is directly related to the decrease of oxygen content in cells,and the hypoxia is an important sign of cancer cells,therefore,the research of nitro reductase has important significance in the field of medicine.In this chapter,we designed and synthesized a fluorescent probe using pyran nitrile as the fluorophore which can dectection the nitro reductase selectively and sensitively,which names NRP-1.In DMSO:PBS = 1:99,pH = 7.4 solvent condition test its spectral properties found that NRP-1 has no fluorescence,after adding nitro reductase and coenzyme NADH,NRP-1 at 537 nm emits strong fluorescence signal.Cell experiment showed that was in hypoxia Hi5 significantly enhanced fluorescence phenomenon,at the same time,NRP-1 was also successfully applied to the detection of tumor hypoxia in mice,shows that NRP-1 is not only the probe specific recognition of nitro reductase,is a highly selective,low oxygen probe with high sensitivity.The design and synthesis of a fluorophore based on anthocyanin as specific,selective recognition of H2S near infrared fluorescent probe compound Cy-S-S,and in DMSO:HEPES = 2:8,solvent pH = 7.2 condition,a series of spectral properties were tested.It is found that with the addition of H2S,UV absorption peak position of Cy-S-S between 700 nm-800 nm is decreases,but a new 400 nm-600 nm absorption peak is enhanced,with a solution color change from green to pink.Meanwhile,the fluorescence enhancement at 633 nm about 300 times can be observed,spectral selectivity experiments show that the Cy-S-S showed a high sensitivity towards H2S and achieved fluorescent recognition.At the same time,Cy-S-S was also successfully applied to Hi5 cells and Caenorhabditis elegans like organisms in vivo.
