Gorgon is an important medicinal and edible aquatic vegetable,which contains a variety of polyphenols,polysaccharides,amino acids and other functional components.Gorgon has nourishing,spleen,kidney,stomach and other TCM theoretical efficacy,as well as anti-oxidative,myocardial ischemia repair effect,and promote gastrointestinal peristalsis,reduce blood pressure,diuretic,inhibit the growth of cancer cells and bacteria.In this paper,polyphenols from gorgon shell and gorgon kernel were extracted,and the composition analysis of gorgon polyphenols,antioxidant activity in vitro and protective effects on oxidative damage of mouse cells were studied in order to provide a theoretical basis for the further development and utilization of gorgon polyphenols.?1?Ultrasound-assisted optimization of gorgon polyphenols extraction process:This experiment was based on single factor Test and the process of ultrasonic assisted extraction of gorgon polyphenols were optimized by response surface method.The response surface analysis model is well fitted with the actual situation,and the optimum extraction condition of gorgon polyphenols is described as,solid-liquid ratio is 1:59?g/mL?,the ultrasonic time is 51 min,the extraction temperature is 50?,the acetone volume fraction is 48%.Under this condition,the gorgon shell polyphenols extract is 127.60 ?g.g-1 FW,The polyphenol extraction amount of gorgon kernel was 10.86 ?g.g-1 FW.?2?HPLC-MS/MS method for gorgon polyphenols composition analysis:The gorgon polyphenols were obtained by ultrasonic assisted extraction,and HPLC-MS/MS was applied to qualitatively and quantitatively detect gorgon polyphenols from shell and kernel.This method successfully detected gorgon shell containing 9 active ingredients,respectively:Hypericum,ferulic acid,kaempferol phenol,celery phenol,caffeic acid,quercetin,celery,rutin,gallic acid.Gorgon kernel contains 9 active ingredients,namely:Ferulic acid,kaempferol phenol,celery phenol,caffeic acid,quercetin,celery,rutin,osmanthus herb,gallic acid.The experimental results show that there is a significant difference in the content and composition of polyphenols in gorgon shell and gorgon kernel.?3?Antioxidant activity of gorgon polyphenols:This experiment was to determine the scavenging rate of DPPH,hydroxyl and O2-anion free radicals rate and reduction and total antioxidant ability in different concentrations of polyphenols extract from gorgon shell.Finally,the distribution law of 11 active ingredient contents in gorgon shell polyphenols and its antioxidant properties were combined.Principal component analysis method was used to analyze its main effect components.The results showed that the antioxidant capacity of gorgon polyphenols was closely related to ferulic acid,gallic acid and parsley.?4?The protective effect of gorgon polyphenols on murine macrophages ANA-1 injury induced by hydrogen peroxide.The results showed that gorgon polyphenols from shell could enhance the resistance of cells to oxidative stress,and could significantly improve cell viability,total antioxidant capacity,GSH content,GSH-Px activity,CAT,SOD and other antioxidant enzyme activities in 400?g/mL treatment,and also significantly reduce MDA content and LDH activity.
