Phthalocyanine?Pc?is a synthetic derivative of porphyrins and belongs to the category of benzoazepines.Due to its 18-electron macrocyclic conjugate system,the maximum absorption of phthalocyanine is significantly red-shifted relative to porphyrin.Pc thus has better photochemical properties than porphyrin.Phthalocyanine has a strong coordination ability and can coordinate with almost all metal elements to form metal phthalocyanine with special colors.The absorption spectra of phthalocyanines can be controlled by introducing corresponding substituents or central metal ions into the periphery of phthalocyanines.The maximum absorption of the phthalocyanine can be red-shifted to the bio-light zone.Phthalocyanine compounds have broad application prospects in the biological diagnosis and treatment.However,due to the strong?-?interaction of the macrocyclic structure,phthalocyanine has poor water solubility,which limits their application in the biological field.In this paper,the water solubility of metal phthalocyanine is improved from two aspects and the complexes were selected to study their application in biomedicine.The specific contents included the following two aspects:1.The water-soluble metal magnesium phthalocyanine and the metal zinc phthalocyanine were successfully prepared and characterized by the phthalonitrile method.The introduction of the peripheral ether chain group increases the water solubility and the basicity of the meso-N.Therefore,the finally obtained metal phthalocyanine has both good water solubility and pH response properties.Due to the protonation,the stability of the phthalocyanine is reduced.It also affects the energy transfer from trilinear phthalocyanine to trilinear oxygen.Eventually leads to a decrease in the singlet oxygen yield of protonated phthalocyanine,which restrict the application in sonodynamic therapy.But this part of the work provides ideas for our work in the future.2.A stable metal iridium phthalocyanine complex is synthesized by a one-step insertion method.In order to improve the water solubility and biocompatibility of the metal iridium phthalocyanine,the water-soluble nanoparticles?IrPcCl2-BSA?were obtained by encapsulation of the metal iridium phthalocyanine by bovine serum albumin?BSA?.The experimental results show that the diameter of the nanoparticles is about 130 nm.It has been found that the metal iridium phthalocyanine complex has the properties of a catalyst and it can catalyze hydrogen peroxide to produce oxygen specifically.Oxygen generation was detected in vitro and in vivo by photoacoustic imaging and laser confocal experiments,which providing strong evidence for nanoparticles to catalyze the production of oxygen by hydrogen peroxide.The results of in vitro and in vivo experiments show that IrPcCl2-BSA has excellent sonodynamic treatment effects.The photoacoustic method was used to detect the changes of hemoglobin?Hb?and oxyhemoglobin?HbO2?in tumor tissues with time,which proved that nanoparticles still had good catalytic effect in the lower concentration of hydrogen peroxide.Oxygen generation catalyzed by nanoparticles improves the hypoxic environment in tumors,provides oxygen source for in vivo sonodynamic dynamic therapy,and enhance the treatment effect.IrPcCl2-BSA nanoparticles have achieved the integration of diagnosis and treatment in vivo and have high application prospects in biomedicine.
