Saline-alkali tolerance rice is one of the important contents of the breeding of rice.This study uses class transcription factor effector nuclease?Transcription activator-like effector nuclease TALEN?genome mediated mutagenesis,with excellent varieties of Japonica Rice Kongyu 131 was improved through the loss of material objects,to enhance its DST gene.Salt resistance properties.The specific results are as follows:1.We successfully constructed a TALEN vector with site directed mutagenesis2.Through agrobacterium mediated transformation of TALEN vector of the DST gene mutation,through the detection of transgenic loci PCR,Southern blot analysis;sequence analysis of DST gene target site urea polyacrylamide gel electrophoresis,a total of 2 strains of DST gene target site fragment missing positive T0 transgenic plants,the positive conversion rate 1.16%.3.The transgenic plants with homozygous,T2 plants on saline alkali stress,the proline content survey,germination rate,germination potential,and saline alkali medium growth,leaf stay green,chlorophyll content,H2O2 content,salt tolerance of the transgenic plants found indeed than wild-type control plants,but also is to produce resistant properties.4.Field planting,different agronomic traits,T2 generation plants and control plants survey results show that,in addition to salt tolerance properties significantly enhanced,T2 plant height,tillering,panicle number,panicle,yield,agronomic traits of grain weight were not changed.
