Morchella has an extremely rich nutrient composition that is a valuable edible and medicinal fungus.This article selected the Morchella conica as the research object from Qing hai.We studied the selection of Morchella conica strains,solid cultures,liquid fermentation processesand the optimization of the cultivation solid substrates.The main results are as follows:1.Screeninof different Morchella conica strains on basal medium: The Morchella conica strainsS,Q,M coming from three Different producing areas were cultured in plate culture and slant culturein.The obtained M strain is most suitable as a test strain by contrast the length of mycelium and the long-term growth rate of hyphae.2.Study on the condition of Solid Medium of Morchella conica: Single-factor test was used to determine the Morchella conicasolid medium formulation: glucose20.0 g,corn flour 20.0 g,peptone 1 g,potassium dihydrogen phosphate 1 g,magnesium sulfate 0.5 g,water 1000 mL.3.Experiment on the optimization of morel liquid medium: we had determined a good liquid fermentation culture technology with better biomass and exopolysaccharide content of Morchella conica through the single factor and orthogonal test.The culture conditions were shaker culture and the optimal medium was glucose 20.0 g,peptone 2.0 g,corn flour 30 g,soybean powder 20 g.4.Study on optimization test of the liquid fermentation culture conditions of Morchella conica: using single-factor and orthogonal experiments,the optimal fermentation conditions for determination of Morchella conica biomass and extracellular polysaccharide content were determined as follows: liquid volume 140 mL,temperature 20 °C,shaker speed 210 r/min,inoculum size 10%,pH = 6,bacterial age 5 days.5.Research on the Culture Conditions of Morel of Morchella conica: The optimum culture conditions for Morchella conica through experiments.Wheat is a solid substrate,the nutrient solution formula is: glucose 20.0 g,peptone 0.5 g,magnesium sulfate 0.5 g,potassium dihydrogen phosphate 1.0 g,water 1000 mL.
