Gut microbita,including symbiosis,probiotics,and pathogenic microorganisms,plays an important role in human health by fighting pathogenic microorganisms.Ulcerative colitis(UC)is a common intestinal nonspecific inflammatory disease with an increasing incidence,and its complications may lead to autoimmune disease even life-threatening.Huangqin Tang(HQT)including skullcap,peonies,liquorice and buckthorn,a traditional Chinese formula chronicled in Shang han lun,has been recognized as an effective drug for UC with anti-inflammatory and antioxidative properties.In recent years,more and more researchers have attached importence to explore the structure and function of intestinal flora along with the development of high-throughput sequencing technologies,metagenomics and bioinformatics analysis technology.Therefore,it is immediate significance to study the relationship between HQT and gut microbita in UC.This study aims to investigate whether HQT ameliorated the dextran sulfate sodium(DSS)-induced UC by changing gut microbita.In the present study,we aimed to examine the effect of HQT on DSS-induced UC in mice.The results showed that HQT(9.1,18.2 g/kg)positively influenced the loss of body weight,disease activity index(DAI),reduced the degree of colon shortening and tissue injury induced by DSS treatment.However it had no effect in mice treated with antibiotics and DSS.In addition,real-time quantative PCR(qRT-PCR)revealed that the levels of tumor necrosis factor-?(TNF-?),interleukin-1 ?(1?),interleukin-6(IL-6),and cyclooxygenase-2(COX-2)were also significantly reduced in the HQT treatment groups compared with the DSS group.For another,Miseq sequencing and operational taxonomic units(OTU)cluster analysis were used to indicate the microbial composition and diversity in three groups.The result of diversity index showed the diversity was increased in the DSS group and DSS+HQT group.Principal component analysis(PCA),principal co-ordinates analysis(PcoA),and LDA Effect Size(LEfSe)analysis indicated an obvious difference in groups.The relative abudance of Lactococeus was raised significantly in DSS+HQT group compared with DSS group.However,the relative abudance of Desulfovibrio and Helicobacter were significantly decreased.In conclusion,we suggested that HQT could ameliorate DSS induced inflammation by regulating the gut microbita community structure.
