The Extraction Process Optimization Of Ginseng Notoginseng Saponins In Feiwei Granules And The Establishment Of HPLC Characteristic Maps

1 ObjectiveStudy on optimization of extraction technology and HPLC feature spectrum and the active of components alignment(ACA)of saponins in Radix Ginseng and Radix Notoginseng and ginseng-notoginseng,so that they can provide a basis for quality control of raw materials in the preaparation process and establishing the quality standards of Feiwei granules.2 Contents and Method2.1 The exhaustive herbs survey according to The Pharmacopoeia standards and methodologies.2.2 The optimization of extraction technology of ginseng-notoginseng with five kinds of saponins' content of extraction yield were selected as assessment indice by L9(34)orthogonal test.The factors include solvent consumption(A,8times,10times,12times),immersion time(B,0.5h,1h,1.5h),extraction time(C,1h,2h,3h)and extraction times(D,once,twice,three times)2.3 Establish the HPLC feature spectrum and analysis the saponin components of Radix Ginseng,Radix Notoginseng,and ginseng-notoginseng and Feiwei granules.3 Results3.1 The raw materials in Feiwei granules are in line with the relevant provisions of The Pharmacopoeia.3.2 The result showed that the optimal extracting condition was A3B2C1D3,which is plus 12 immersions in water 1 hour,extraction 3 times,each time 1hour.Under this condition the total content of five saponins is up to 223mg.3.3 There are 8 peaks in HPLC feature spectrum of Radix Ginseng decoction,10 peaks n HPLC feature spectrum of Radix Notoginseng decoction,13 peaks in HPLC feature spectrum of ginseng-notoginseng decoction,and 12-13 peaks in HPLC feature spectrum of Feiwei granules,results showed that they are special.And in ginseng decoction,the ACA of ginsenosidesRg1:ginsenosidesRe:ginsenosidesRf:ginsenosidesRb1 is 8.488:1.641:1:4.308.In notoginseng decoction,the ACA of notoginsenosideR1:ginsenosidesRg1:ginsenosidesRe:ginsenosidesRb1 is 1.307:11.931:1:6.380.In ginseng-notoginseng decoction,the ACA of notoginsenosideRi:ginsenosidesRg1:ginsenosidesRe:ginsenosidesRf:ginsenosidesRbi is 4.807:47.917:4.942:1:23.257.4 Conclusions4.1 Saponins are the master ingredient in Radix Ginseng and Radix Notoginseng and Feiwei granunles,their content can affect the quality and efficacy of this durg.The optimal extracting condition is plus 12 immersions in water 1 hour,extraction 3 times,each time 1 hour.That is higher than other conditions.4.2 The HPLC feature spectrum of ACA of saponins in ginseng-notoginseng has exclusive,and the content of notoginsenosideRi:ginsenosidesRg1:ginsenosidesRe:ginsenosidesRf:ginsenosides Rbi is 4.962:31.077:4.173:1:15.019,which can be used for Feiwei granules quality standard and production quality control.
