This research focuses on the quality control of traditional Chinese compound and Chinese herbal material. It includes two parts:Part I:Chemieal Analysis of Traditional Chinese Medicine Prescription. The compound is composed of Radix et Rhizoma Ginseng, Radix Polygoni Multiflori, Radix Angelicae Sinensis and Rhizoma Chuanxiong, which have the effective constituents of Ginsenoside Rb1, Ginsenoside Rg1, 2,3,5,4'-Tetrahydroxy stilbene-2-O-β-D-glucoside, ferulaic acid and Ligustilide. The main research and its results were reported as follows:A method for detemination of 2,3,5,4'-Tetrahydroxy stilbene-2-O-β-D-glucoside, ferulaic acid and Ligustilide in Chinese Medicine Prescription by HPLC was established. The linear relation was excellent for determination on C18 column with acetonitril-0.15% Phosphoric acid as mobile phase. The gradient elution mode was applied in chromatographic separation. The detection wavelength was at 320nm, the flow rate was 1.0ml·min-1 and column temperature was 30℃. The method is simple, precise and reproduciable and can be used for determination of 2,3,5,4'-Tetrahydroxy stilbene-2-O-β-D-glucoside, ferulaic acid and Ligustilide simultaneously in Chinese Medicine Prescription effectively. In this paper, the method is optimized to extract Ginsenoside Rb1 and Ginsenoside Rg1 from Chinese Medicine Prescription. Ultrasonic waves and heating circumfluence were discussed, and the extraction effect was compared. Ultrasonic extraction has significant advantages in fast and high efficiency. The method, which is capable of detecting both critical and common components simultaneously from multiple raw materials, is also an innovation.PartⅡ:Comparison of Salicin and Rutin Content in Propolis with Poplar Leaves and Buds. The HPLC method for determination of Salicin and Rutin in Propolis was developed. The separation of the two components was achieved on a reversed-phase Kromasil C18 column with methanol-0.15% Phosphoric acid gradient elution at a flow rate of 1.0 mL·min-1, the detection wavelength was at 268nm. And this method is simple, accurate and reliable by evaluating the method validation data, and can be used for the determination of Salicin and Rutin in poplar leaves and buds. Comparison of Salicin and Rutin content in propolis, fake-propolis, propolis-extract, poplar leaves and buds were determined by HPLC. As the result, the two glycosides do not appear in propolis, fake-propolis and propolis-extract. In a previous summary, the author firstly proposed an opinion that glycosides does not exist in original propolis. Experimental data in this paper supported the opinion for the first time. Therefore, it is suggested the relevant standards in use to be reconsidered accordingly.
