This study based on the National Natural Science Fund project "Yiqi huoxue bioactive component inhibit diabetic Hamster cardiomyopathy and ventricular remodeling and apoptosis mechanism"(No.81173258).Includes".Based on the study of the total use of Danshen Yin and Sheng Mai San on myocardial fibrosis in diabetic cardiomyopathy in rats,it conducts a research on the Yiqi huoxue bioactive component which made up of Danshen Yin and Sheng Mai San optimally.The study shows that the deficiency of both qi and yin is the basic syndrome of DCM,while the blood stasis is the most accompanied symptoms and signs.So the "Yiqi yangyin and Huoxue huayu" is the basic treatment.Besides the research of the treatment of the Diabetic cardiovascular disease by the total use of the Sheng Mai San and Dan Shen Yin is in a majority and there are various dosage forms of the effective monomer component of the traditional Chinese medicine among them in the clinical application which makes the study of drug effect and pharmacokinetics clearer to the benefit of the study of modernization of Chinese Traditional Medicine.Then the uni form design and methods,suitable for multi-factor and multilevel trial study,have an increasing number of the study of the combination of the effective component of the traditional Chinese medicine with some success.Object:The diabetic cardiomyopathy(DCM)is the pathological changes of specificity in a long-term hyperglycemic condition.Based on the early clinic and the effective experimental study of the total use of Shen Mai San and Dan Shen Yin,it conducts the further research about the effective component of the traditional Chinese medicine,which shows the protection of the effective component on myocardial fibrosis and fibroblast(CFs)in diabetic cardiomyopathy in rats.It explores the mechanism of action of the total use of Sheng Mai San and Danshen Yin for DCM,adopts the uniform design and methods and finds the best compatibility and the regularity in drug combination for on myocardial fibrosis in diabetic cardiomyopathy.Method:Taking the known six effective component(total Ginsenoside Ginseng Root?ophiopogonpolysaccharide?Edible oil?tanshinone?A?Sandalwood Oil?bomyl acetate)from the Chinese herbal compound prescription of the Sheng Mai San and Dan Shen Yin on myocardial fibrosis as research objects,it uses the model of streptozotocin(STZ)to induce SD rats to get the myocardial fibrosis in diabetic cardiomyopathy and adopts the uniform design and the form of the six factors and the twelve levels to get the best dispensation by regression analysis step-by-step with the index of the ventricular remodeling and the myocardial apoptosis of the rats as a screen.After four weeks of dosing the remedial medicine for groups,we take out of the hearts of the rats,keep the eyes on the form and frame of myocardium with HE stain and myocardial collagen type ?and ? with the Picric acid-Sirius red staining and the content of collagen type ? and ? with enzyme linked immunosorbent assay after anesthetizing.To determine the index of the apoptosis,we use Terminal-deoxynucleoitidyl Transferase Mediated Nick End Labeling(TUNEL)of inducing the Deoxyribonucleic acid(DNA)terminal transferees,while examining the apoptosis gene Bax,Bcl-2,the specific value of them and the expression of the protein P53by Westenblot,Therefore we can get a prognosis of the best dose proportion by the regression equation from the above index.Results:(1)By observing the mass index of the heart of the model set on myocardial fibrosis in rats,myocardial collagen index,apoptosis index and the myocardial morphology,we compare the model and normal group,find the difference with statistical significance(P<0.05)and conform to the pathological feature of DCM,which means a successful model.(2)From the A dispensing screening from Myocardial content of type I collagen,we can get the regression equation:Y=26.704-0.01BF-2.05E-0.019BC,which reminders sandalwood oil a larger and negative Coefficient,inhibiting the type? collagen hyperplasia and playing a main role.Besides,ophiopogonpolysaccharide,edible oil and bomyl acetate play a synergistic role to inhibit the collagen hyperplasia together.When the ophiopogonpolysaccharide gets 300mg,Edible oil gets 1mg,Sandalwood Oil gets 10mg and bomyl acetate gets 10mg,the collagen hyperplasia has the minimum value.(3)With the tape ? collagen content as the dependent variable,we find the multivariate linear regression,Y=6.188-0.003BE+0.057D+3.023C-0.069CD,which reminders that Tanshinone ?A and sandalwood oil play a main and synergistic role to inhibit the tape ? collagen hyperplasia together and ophiopogonpolysaccharide and sandalwood oil get a interplay to inhibit the collagen hyperplasia.When the ophiopogonpolysaccharide gets 300mg,Sandalwood Oil gets 10mg,Edible oil gets 0.25mg,and Tanshinone ?A gets 5mg,it gets the best ratio.(4)With the AI as the dependent variable,we find the multivariate linear regression,Y=0.663-0.018A+0.0001EF-0.224C,where the quotient of A?C is negative in the equation.When the total Ginsenoside Ginseng Root gets 200 mg,Edible oil gets 1mg,Sandalwood Oil gets Omg,and bomyl acetate gets Omg,it gets the best ratio.So the evaluation of both of A and C has the negative correlation with AI while the Sandalwood Oil and bomyl acetate have the positive correlation.(5)With the specific value of Bcl-2/Bax as the dependent variable,we find the multivariate linear regression,Y=0.276-0.004D+1.05*E-4BE,where the interaction of above two plays a regulating effect for the expression of Bcl-2 and Bax while the tanshinone?A don' t.And when the ophiopogonpolysaccharide gets300 mg,Sandalwood Oil gets 10mg,it gets the best ratio.Conclusion:(1)The combination of the component of the six kinds of traditional Chinese medicine above can play an effective role on the index of the myocardial fibrosis in diabetic cardiomyopathy in rats.When in the different pathological mechanism,the different medicine playing an important role should be different.(2)It adopts the uniform design and methods to study the pattern of the combination of the component of.the traditional Chinese medicine,which provides a suitable approach for the research about the best compound combination to screen the against Diabetic myocardial fibrosis from the effective component of the traditional Chinese medicine.
