| Objective:To investigate the effects of Survivin-RNA Gene Silencing Combined with SanWuBaiSan on Apoptosis and Migration of Gastric Cancer Cell Line SGC-7901.Methods:1.SGC-7901 cells were divided into three groups: CON group(Control group,Blank cell group)、 NC group(Negative Control,Negative Control silencing lentivirus transfection cell group)、 KD group(Knock Down,Survivin gene silencing lentivirus transfection cell group),The groups were treated by lentivirus transfection,after transfection for 72 hours,The transfection effect was observed under fluorescence microscope.2.To investigate the effects of lentivirus transfection on the apoptosis and migration of SGC-7901 cells:Experimental group: CON group、 NC group 、 KD group,The apoptosis of each group was detected by flow cytometry,The migration ability of each group was detected by scratch test.3.To investigate the effects of lentiviral transfection combined with SanWuBaiSan on the apoptosis and migration of SGC-7901 cells:Experimental group: CON+traditional Chinese medicine group 、 NC+traditional Chinese medicine group 、 KD+traditional Chinese medicine group,The apoptosis of each group was detected by flow cytometry,The migration ability of each group was detected by scratch test.Results:1.After transfection for 72 hours,green fluorescence was observed in KD group and NC group,and no fluorescence was observed in CON group.2.The apoptosis rate of KD group(5.47%)was compared with that of CON Group(1.283%)and NC group(1.880%),The difference was statistically significant(P<O.01);The 24 h migration rate of KD group(27.80%)was compared with that of CON group(59.00%)and NC group(51.60%),The difference was statistically significant(P<O.01).3.The apoptosis rate of KD+traditional Chinese medicine group(30.52%)wascompared with that of CON+traditional Chinese medicine group(8.49%)and NC+traditional Chinese medicine group(9.38%),The difference was statistically significant(P<O.05);The 24 h migration rate of KD+traditional Chinese medicine group(17.60%)was compared with that of CON+traditional Chinese medicine group(48.40%)and NC+traditional Chinese medicine group(44.60%),The difference was statistically significant(P<O.05).Conclusions:1.Survivin-RNA gene silencing can induce the apoptosis of gastric cancer SGC-7901 cells and inhibit the migration of SGC-7901 cells.2.Survivin-RNA gene silencing combined with SanWuBaiSan can induce the apoptosis of gastric cancer SGC-7901 cells and inhibit the migration of SGC-7901 cells.The results of this study show that Survivin-RNA gene silencing combined with SanWuBaiSan can induce the apoptosis of gastric cancer SGC-7901 cells and inhibit the migration of SGC-7901 cells.The mechanism may be related to the inhibition of the expression of Survivin by SanWuBaiSan,and the Survivin gene can inhibit cell apoptosis and participate in angiogenesis. |
