Vascular tissue engineering is the use of the normal cells of blood vessel walls and biodegradable materials preparation,reconstruction and regeneration of blood vessels to replace of material science,also known as the "regenerative medicine".It is a use of biological active substance itself,the culture outside the body,or the building method,reconstruction or repair of organs and tissues.Tissue engineering research is the core of scaffold materials,applied to the support of clinical material must have good blood compatibility and non-toxic side effects.Decellularized scaffold material completely made up of natural extracellular matrix,which can be used to maintain the mechanical properties of blood vessels,but because of its structural defects,decellularized scaffold cannot directly as a stent materials used to construct tissue engineering vessels,decellularized scaffolds for polymer film coating processing is the important way to solve the above problem.PLA(Polylactic acid,PLA)as a new type of green polymer material,with good mechanical properties and biodegradability,it is able to active as artificial blood vessels,artificial hearts and other material in the field of medicine.Itself the biocompatibility of poly(lactic acid)(PLA),however,it is still cannot meet the standards of medical materials,therefore,the people working on surface modification of poly(lactic acid)(PLA)to make the effects on the human body,reduce or avoid thrombosis after have better anticoagulant hemorrhagic and compatibility.Zwitterions of phosphorycholine belong to the phosphoryl choline compounds,the hydrophobic part is long carbon chain nonpolar hydroxyl,on the other side as part of hydrophilic bile bases phosphate group,so that the ammonium phosphate amphoteric ion modified materials have better blood compatibility.Therefore,ammonium phosphate amphoteric ion can be used as the surface modification of biomaterials material,to improve the tissue compatibility and blood compatibility of medical material.This paper with phosphorus trichloride and ethylene glycol as the starting material,the preparation of zwitterions of phosphorycholine,with nuclear magnetic resonance(NMR),infrared spectrum(FT-IR)on the synthesis of ammonium phosphate ions for structural characterization,testing its chemical structure.This paper,by using chemical method of anionic surfactant sodium dodecyl sulfate and nonionic surfactant Triton X-100 synergy,in as much as possible to keepSecond,and then through chemical grafting method for the preparation of zwitterions of phosphorycholine of polylactic acid/decellularized scaffold modified,improve its blood compatibility.This paper also by X-ray photoelectron spectroscopy(XPS)analysis,thermogravimetric analysis(TGA),the static contact Angle(SCA),tensile properties test study on synthesis of ammonium phosphate amphoteric ion/polylactic acid membrane/decellularized scaffold had been used to characterize the morphology and structure;With hemolysis experiment,experiment,blood clotting time in vitro,dynamic blood coagulation,platelet adhesion experiments,cell toxicity experiment,by means of the growth of endothelial progenitor cells adhesion experiment blood and cell compatibility was studied.Finally the above vascular patches were implanted in vivo experiments on animals.B to exceed the experimental results showed that in the case of without the use of anticoagulants,blood can flow smoothly in vascular patch embedded parts.
