Objective:To observe the effects of phospholipase C(PLC)and inositol 1,4,5-trisphosphate receptor(IP3R)on LI-RI process and skeletal muscle tissue of rats after pretreatment with Taohong Siwuye.Dynamic changes,further clarify the pathological mechani m of LI-RI from the molecular level and explore the molecularbasis of Taohong Siwuye in preventing and treating this disease.Methods:78 male SD rats were randomly divided into 4 groups:normal group,ischemia-reperfusion group(IR group),Taohong Siwuye group(T-IR group)and U73122 group(U-IR group).Blood reperfusion group,Taohong Siwuye group and U73122 group were divided into 0h,2h,4h,and 8h groups at different time points after reperfusion.HE staining was used to observe the skeletal muscle morphology;RT-PCR was used to detect the expression of PLC and IP3R genes in skeletal muscle tissue;finally,the data were statistically analyzed and compared.Results:The results of HE staining showed that,in addition to the normal group,rats of IR,T-IR,U-IR all showed different degrees of muscle fiber structure abnormalities,such as muscle fiber swelling,irregular arrangement,widening of crevices of muscle fiber,etc.,among which IR group The most obvious;compared with the normal group,IR,T-IR,U-IR group skeletal muscle tissue PLC,IP3R mRNA expression levels have increased(P<0.05);and T-IR,U-IR The levels of TNF-?,IL-1?,and the corresponding PLC,IP3R mRNA levels at the same time point were lower than that of IR group(P<0.05).Conclusion:1.The high expression of PLC and IP3R may be involved in the process of pathological signal transduction of rat skeletal muscle injury after IR.2.Taohong Siwuye can reduce the levels of PLC and IP3R in rat skeletal muscle cells after IR,so as to reduce the severity of LI-RI.
