Objective To investigate the two drugs: methotrexate(MTX) andalkylating agent(chlorethazine, HN2) if they have the synergistic effect or notwhen they were used unitedly which can inhibit the Henrietta Lacks strain ofcancer cells(Hela cells) from growing in vitrocolture environment. Furthermore,try to find out the rudimentary mechanism of the role and to provide scientificbasis for the clinic medication. Method Using the MTT method to measure the suck-light degree valueafter using the MTX and HN2 alone or combined; using the flow cytometry withPI staining to detect the changes of cell cycle and apoptosis occurred in the Helacells after using the MTX and HN2 alone or combined. Result The drugs of MTX and HN2 all can inhibit the growth ofthe Hela cells and the trend is more obvious when the dosage is increased, wecan find the cellar inhibitory rate increases, the proliferation index numberdecreases and the MTX can cause many changes in cell cycle; when they wereused unitedly, the effect is stronger than they were used alone and the trend isalso more obvious when the dosage is increased, the cellar inhibitory rate, theproliferation index number and the change in cell cycle took on differentsituations. We got the p value is less than 0.05,so we could consider the twodrugs used unitedly have the synergistic effect. Conclusion MTX is the specific depressant of cell cycle and it can makethe cell cycle change and a few part occurs the apoptosis. But the HN2 is theCCNSA(cell cycle non-specific agent), the inhibition to the cell manifested itcan cause cell death. When the two drugs were combined to use, the synergisticeffect is showed because they could act in different period and various site andtarget.
