| The diterpenoids,alkaloids and triterpenoids in tripterygium glycosides are the main active ingredients,and the most representative active ingredient of diterpenoids is triptolide(triprolactone alcohol)and triterpenoids.The most representative active ingredient is tripterine,although researchers have conducted more comprehensive and in-depth research on it for decades.However,the extraction process of tripterygium glycosides was studied only for a single triptolide or tripterine as an indicator component,failing to combine several active ingredients for the extraction process.In addition,the quality control standards for Tripterygium wilfordii still remain in the study of fingerprints or only the content of active ingredients,failing to combine its main active ingredient and fingerprint,which is not enough to explain the best herbs of Tripterygium wilfordii.The main purpose of this dissertation is to explore the optimal extraction process of Tripterygium glycosides using multiple active ingredients in Tripterygium glycosides as an index,so as to fully reflect the optimal extraction process through the content of multiple active ingredients,and to optimize its extraction process.At the same time,this paper also studied the inspection of different Tripterygium wilfordii pieces and different Tripterygium wilfordii medicinal materials,explored and analyzed the optimal chromatographic conditions for the four active ingredients of Tripterygium wilfordii,and analyzed the quality of the four main active ingredients of different Tripterygium wilfordii medicinal materials.Therefore,this paper reports on the research work from the following four aspects.Firstly,the research progress related to the study of Tripterygium wilfordii medicinal materials is mainly reviewed in the preface,and the research purpose and significance of this thesis topic were described.Secondly,the moisture content,ash content and acid-insoluble ash content of Tripterygium wilfordii slice and Tripterygium wilfordii from different places of origin were examined.The differences in the moisture content of different Tripterygium wilfordii slice and Tripterygium wilfordii extract are analyzed,and the difference between them is shown.Ash content(Inorganic ingredients such as inorganic salts and oxides)differences in content,acid-insoluble ash(sediment and mechanical substances).Thirdly,the quality of Tripterygium wilfordii from different producing areas was analyzed by high performance liquid chromatography(HPLC),mainly from the four major producing areas of Guizhou,Sichuan,Yunnan and Jilin.The same high performance liquid chromatographic conditions were used to detect the Tripterygium wilfordii from different producing areas.The content of the four active ingredients,and the sum of its four active ingredient content,with the highest total content of the four active ingredients is the best medicinal material.Finally,the best medicinal materials were used to study the extraction process of Tripterygium glycosides,and the best extraction process was determined by high performance liquid chromatography analysis: it was heated and refluxed with 95% ethanol water bath to extract 5 times.Each time 3.5 h,the material to liquid ratio of 1:6;followed in accordance with the company’s process is unchanged,adding 30 mL of ethyl acetate ultrasonic extraction,with 3.5 g neutral alumina and 1cm endoscope column loading,with 35 mL acetate B Elution of esters.Chapter 5 summarizes and looks forward to each chapter. |
