Tripterygium Wilfordii Hook is the root, leaf and flower of celastraceae plantsTripterygium Wilfordii. It mainly used in Rheumatoid Arthritis, Lupus Erythematosusand some other autoimmune diseases. It also used for Anti-organ rejection effect inclinical. This paper set up high performance liquid chromatograph (HPLC) testsystem for3active components in different parts of Tripterygium Wilfordii Hookwhich are from7different provinces,34different producing areas,in order to selecthigh quality source.The main content is as follows:1. Set up high performance liquid chromatograph (HPLC) test system forDiterpenes, triterpene of Tripterygium Wilfordii Hook, and Screening mobile phasefor Triptolide, Wilforlide A,Tripterine. The result is that mobile phase for Triptolide iswater: methanol=63:37(v/v); gradient elution:0â†'44min,methanol(37%)ï¼›45â†'55min,methano(l100%)ï¼›55.1â†'63min,methano(l37%)ï¼›velocity:0.25ml/minï¼›column temperature:30℃;UV:220nmï¼›the retention time of triterpene is14min ofthis test system. The mobile phase for wilforlide A, Tripterine is water: methanol=18:82(v/v); gradient elution:0â†'20min,methanol(82%)ï¼›21â†'30min,methanol(100%)ï¼›30.1â†'38min,methano(l82%)ï¼›velocity:0.25ml/minï¼›column temperature:30℃;UV for wilforlide A:202nmï¼›UV for Tripterine:425nm.2. Set up HPLC test system for the distribution of Triptolide in leaf and rootof Tripterygium Wilfordii Hook. Sample size of Triptolide shows a good linearrelationship in the range of11.28ng-564ng. The analysis result indicates that thecontent of Triptolide is difference among Tripterygium Wilfordii Hookfrom differentproducing area, the highest Triptolide is7.255ug/g in pingxiang of Jiangxi, the lowestis0.139μg/g in Jiangkeng of Fujian. The average is2.336ug/g. the content ofTriptolide in leaf and root is difference though from the same producing area. Ingeneral, the content of Triptolide in root is higher than in leaf. The highest Triptolidein root is6.337ug/g from wuyi moutain of Fujian, The highest Triptolide in leaf is3.486ug/g from Taining of Fujian, The content of Triptolide in root is higher twotimes than that in leaf. 3. Set up HPLC test system for the distribution of Tripterine in leaf and root ofTripterygium Wilfordii Hook. Sample size of Triptolide shows a good linearrelationship in the range of243.5ng-9740ng. The analysis result indicates that thecontent of Wilforlide A is hardly to detect of this test system for most of samplesin leaf. However, The content of Wilforlide A in root is higher than that in leaf. Theaverage is3.206μg/g. The content of Wilforlide A is different between differentsample from different producing area The highest Wilforlide A in root is17.33μg/g(Datian of Fujian), the lowest is0.472μg/g(Leigong of guizhou). All thesample can detect the content of Wilforlide A.It can domesticate and improve thesample from Dabu of Fujian, Licang of Yunan,Wuyi of Zhejiang in order toextracting Wilforlide A in leaf.4. Set up HPLC test system for the distribution of Tripterine in leaf and root ofTripterygium Wilfordii Hook. Sample size of Tripterine shows a good linearrelationship in the range of108.5ng-1302ng. The analysis result indicates that thecontent of Tripterine is hardly to detect of this test system for most of samples inleaf. However, The content of Tripterine in root is more higher than that in leaf. Theaverage is8360.12μg/g. The content of Tripterine is different between different samplefrom different producing area The highest Tripterine in root is18882.1μg/g in Ba xianshi of Fujian, the lowest is2832.89μg/g in Chang de of Hunan. All the sample candetect the content of Tripterine. The result of the content of Tripterine is consistent with therelated researchs.5. Use EXCEL and DPS to do the relativity analyzing, the result shows therelationship of distributing of same components from the different parts is similar. Thecoefficient of triptolide distributing from leaf and root is0.249, The coefficient ofwilforlide A is0.291. The coefficient of Tripterine. is0.204.The relativity coefficient ofdistributing of different components from the same part is different. The relativity oftriptolide and Tripterine in the root is the biggest, the coefficient is0.30. The relativitycoefficient of different components in leaf is hardly existed.Then5high qualitysamples were selected:Baxianshi of Fujian,Datian of Fujian,Songyang ofZhejiang,Wuyi moutain of Fujian,Pingxiang of Jiangxi. 6. Study on leaf and root biomass from8-yr-old Tripterygium Wilfordii Hook, Byanalyzing the relation of biomass between leaf and root, to provide scientific basis forthe rational utilization of leaf resource. The analysis result indicates that the highestyield of leaf is1.52Kg each plant,the lowest is0.84Kg each plant from8-yr-old plant.the average is1.07Kg, The drying rate of leaf is36%. the highest yield of root is3.01Kg each plant,the lowest is1.75Kg each plant.the average is2.13Kg, The dryingrate of leaf is53%.The weight ratio of leaf to root is1:2when they are fresh, Theweight ratio of leaf to root is1:3when they are dry. Use EXCEL to do the relativityanalyzing, the result shows that the relativity of leaf and root is big, the coefficientcoefficient is0.905when they fresh and the coefficient coefficient is0.606when theydry.
