Objective: To compare the levels and proliferation of EPCs between different d oses of statins taken by unstable angina peri-percutaneous coronary intervention,to e xplore the mechanism of the benefit of high loading dose Statin treatment before PC I.Methods: 128 patients with unstable angina were randomly divided into either r ecieving 20 mg rosuvastatin or 10 mg started from 1 day before PCI.All patients had no statins in the past 3 months.Both groups started taking rosuvastatin 1 day before PCI.According to the results of coronary angiography,30 patients in each group w ere selected according to the exclusion criteria.Blood samples were collected at basel ine,24 h and 7d after PCI.Then these blood samples were isolated,cultured and ide ntified,in order to count the number of EPCs,while the proliferation and migration of EPCs were determined by MTT assay.Measure migration function by Wound Healin g.Results: 11 days after the separation of EPCs,high-dose statin-treated group has more cell colonies and stronger adhesion.Wound Healing result display that the migr ation speeds of the two groups of cells were both faster and there was no significant difference between the two groups.Gradual healing of cells gradually occurs after 24 hours of cell isolation and culture.Significant differences were obserbed among gr oups as regard to the levels of EPCs 24h(88.90±7.230 vs 85.10±6.567)and 7d(79.07±8.542 vs 70.03±9.514)after PCI.The levels of migration and OD were higher in 20 mg group only documented in 7d after PCI(46.67±5.990 vs 42.53±6.653,0.789±0.072 vs 0.719±0.091,p<0.05).Conclusion: High loading dose treatment(20mg rosuvastatin)in perioperative per iod of PCI may benefit from its enhancement of levels,proliferationg and migration o f EPCs.By positively affecting the number and function of EPCs,this may be one o f the beneficial mechanisms for perioperative intensive statin therapy.
