Lung cancer ranks first in the incidence and mortality of various malignant tumor diseases,and the incidence of lung adenocarcinoma in lung cancer is the highest.The present study shows that the phenothiazine antipsychotic chlorpromazine has inhibitory effect on glioma,liver cancer,etc.but chlorpromazine has inhibition effect on lung adenocarcinoma and mechanism of action is still without a large number of reports.Objective: To determine whether phenothiazine antipsychotics chlorpromazine has inhibitory effects on lung adenocarcinoma A549 cells and its mechanism of action.METHODS: Lung adenocarcinoma A549 cells were routinely cultured.The effects of different concentrations of chlorpromazine on the growth ability,colony formation ability,migration and invasion of lung adenocarcinoma A549 cells were observed.Protein expression in lung adenocarcinoma A549 cells after drug treatment was tested by western blot.Experimental results: MTT results showed that the antipsychotic drug chlorpromazine can significantly inhibit the proliferation and growth of lung adenocarcinoma A549 cells.Plate colony formation assays showed that chlorpromazine had a significant inhibitory effect on the colony forming ability of lung adenocarcinoma A549 cells.Transwell migration and invasion experiments showed that the antipsychotic drug chlorpromazine has a good ability to inhibit the migration and invasion of lung adenocarcinoma A549 cells.Western blot experiments showed that chlorpromazine inhibited lung cancer A549 cells through CaMKII/c-fos pathway.The above inhibitory effects were dose-dependent.Conclusion: The phenothiazine antipsychotics chlorpromazine have inhibitory effects on lung adenocarcinoma A549 cells.
