| Objective:To observe the role of endoplasmic reticulum stress-apoptosis pathway in rat Achilles tendinopathy,and to detect the expression of apoptosis index and endoplasmic reticulum stress related proteins GRP78,eIF2α and CHOP in rat Achilles tendinopathy To explore the possible mechanism of the occurrence of Achilles tendinopathy in rats and the prevention and treatment effect of Zheng’s No.1 new wound medicine on rat Achilles tendinopathy.Methods:96 8-week-old SPF SD rats were randomly divided into blank control group(C group),simple jumping group(J group),simple dressing group(M group)and jumping dressing group(JM group).The group is divided into 4W,6W,8W groups according to the time of material selection,8 in each group.The J group and the JM group were subjected to electrical stimulation jump modeling.The M group and the JM group began external application of Zheng’s No.1 new wound medicine intervention,observed the changes of vital signs in rats,and recorded the number of effective jumps in rats.They were taken at the 4th,6th,and 8W weekends.The morphological changes of Achilles tendon were observed,the apoptosis index was detected by Tunel method,and the expression of GRP78,eIF2α and CHOP protein in Achilles tendon tissues were detected by Western Blot.Data processing was performed using Graphpad Prism 5 software,and one-way variance and independent sample T test were used for index evaluation.Results:(1)Number of effective hopping results: The number of effective jumps in the J group and the JM group showed a downward trend.In the first 5 weeks,the number of effective jumps in J group and JM group was basically the same,there was no significant difference(P>0.05).The number of effective jumps in JM group was significantly better than that in J group at 6 weeks(P<0.05).(2)HE staining results: The morphological structure of Achilles tendon tissue was basically the same at the 4th,6th and 8th week of the rats in group C and M.The collagen fibers were not disordered and the morphological structure of chondrocytes was normal.The rats in group J were seen at the 4th week.The collagen fibers in the sputum were slightly less regular and the structure was slightly loose.At the 6th week,obvious pathological changes appeared,and some collagen fiber rupture,inflammatory cell infiltration and new capillary growth were observed.At the 8th week,the sputum fibers were sparsely arranged.Large-area fiber rupture;after the external application of Zheng’s No.1 new wound medicine intervention,the morphological structure of Achilles tendon tissue in JM group was significantly different from that in J group,and the pathological changes of Achilles tendon tissue were significantly improved.(3)TUNEL apoptotic results: There was no significant difference in the apoptotic index of Achilles tendon tissue between group C and M group.The apoptosis index of group J was significantly increased,and in group 4,6,and 8 and group C,There were significant or very significant differences in M group and JM group.After the intervention of Zheng’s No.1 new wound medicine,the apoptosis index of JM group was significantly lower than that of J group,and there was no significant difference between group C and M group.(4)Western Blot results: The expression of GRP78 protein in Achilles tendon tissue of group J increased significantly after the 4th week,reached the highest peak at 6 weeks,and then showed a slow downward trend;eIF2α showed a sharp increase after 4 weeks,and with group C There was a significant difference(P<0.01),and it showed a slow downward trend after 6 weeks.The expression of CHOP protein continued to increase from the 4th week,and the expression level of the CHOP protein was significantly different from that of the C group after 8 weeks(P<0.05).After the intervention of Zheng’s No.1 new wound medicine,the expression of GRP78,eIF2α and CHOP protein in the Achilles tendon tissue of JM group was down-regulated,and decreased compared with J group,and C group and M group.There were no significant differences.Conclusions:(1)Zheng’s No.1 new wound medicine intervention has a certain preventive effect on the formation of Achilles tendinopathy in rats.(2)Endoplasmic reticulum stress and its mediated apoptosis are related to the occurrence of achilles tendinopathy in rats.ERS mediates the apoptosis process of rat Achilles tendinopathy,which may be one of the pathogenesis of Achilles tendinopathy.(3)External application of Zheng’s No.1 new wound medicine can significantly down-regulate the expression of endoplasmic reticulum stress-related proteins GRP78,eIF2α and CHOP in rat Achilles tendon tissue,and effectively regulate the endoplasmic reticulum stress-apoptotic response in rat Achilles tendon tissue and inhibit sputum Apoptosis. |
