Objective : To study the prevention and treatment of Zheng's No.1 New Wound Medicine on rat Achilles Tendinopathy model by observing the morphological changes of achilles tendon tissue during the experiment;testing the expression of Mechano-Growth Factor and endothelium Nitric Oxide Synthase as a force sensitive factor,to explore them changes in the development of tendinopathy and the effect of Zheng's No.1 New Wound Medicine.Methods:Ninety-six male Sprague-Dawley rats were randomly divided into control group,medicine group,jumping group and jumping medicine group,with 24 rats in each group.Each group of rats was divided into 4-weeks group,6-weeks group and8-weeks group according to different sampling time,8 in each group.Group C was fed with normal diet.Group M was fed with normal diet and then treated with Zheng's No.1 New Wound Medicine from the 4th week.Group J was treated with Electric Stimulation Jumping Method after adaptive feeding.Group JM was treated with Zheng's No.1 New Wound Medicine from the 4th week in addition to Electric Stimulation Jumping Method.Observe and record the general condition indicators of rats.Achilles tendon tissues were taken 24 hours after the end of modeling at 4th,6th and 8th week.The morphological changes of the tissues were observed by HE staining.The expression of mechanical growth factor and endothelial nitric oxide synthase was detected by Western Blot method.The data were analyzed using independent sample T test and two-way analysis of variance.Results:1.In terms of jumping ability,the number of effective jumps in group J and group JM showed a downward trend.The number of effective jumps in group J and group JM was slightly different from the 1st week to the 5th week,but no significant difference was observed.(P>0.05);the number of effective jumps in group J wassignificantly less than that in group JM from 6th week to 8th week(P<0.05).2.In terms of morphological observation,the tendon fibers in group C and group M were wavy and curved,and the arrangement of tendon fibers was close,and there was no significant change in the 4th,6th and 8th weeks.At the 4th week of group J,the tendon fibers were arranged irregularly,a few fibers were broken,and the gap was slightly enlarged.At the 6th week,the tendon fibers were arranged disorderly,some fibers were broken,the number of nuclei increased,inflammatory infiltration and vascular growth were seen locally.At the 8th week,the tendon fibers were broken in a large area,the gap was enlarged,the number of nuclei increased significantly,and the local vascular growth was observed.In JM group,tendon fibers were relatively regular and occasionally slightly enlarged at the 4th week,local tendon fibers were irregularly arranged and a few fibers were broken at the 6th week,and tendon fibers were disorderly arranged at the 8th week,some fibers were broken and a few inflammatory cells were seen locally.3.In terms of MGF protein expression,there was no significant difference in the expression of group C and group M,but the expression of MGF in group J showed an upward trend,while that in group JM decreased first and then increased.MGF protein expression in group J was always the highest in each stage,followed by group JM.At week 4,there was no significant difference in MGF protein expression between groups(P > 0.05);at week 6,there was significant difference between group J and other groups(P < 0.05),group JM and other groups(P < 0.05).At the 8th week,there were significant differences between the control group(group C,group M)and the model group(group J,group JM)(P < 0.05),but there was no significant difference between group J and group JM(P > 0.05).4.In terms of eNOS protein expression,there was no significant difference in the expression between group C,group M and group J at each stage,and group JM showed an upward trend,while group JM showed a downward trend.The expressionof eNOS protein in group J was always the h ighest in each stage.At week 4,there was significant difference between group J and group C and group M(P < 0.05),and there was no significant difference between group JM and other groups(P > 0.05).At week 6,only group J and group C existed.At the 8th week,there was significant difference between group J and other groups(P < 0.05),but group JM was only significant with group J(P < 0.05).Conclusion:1.According to the morphological results,Zheng's No.1 New Wound Medicine has preventive and therapeutic effects on Achilles Tendinopathy in rats.2.Its control effect may be achieved by inhibiting the abnormal increase of nitric oxide synthase and the excessive synthesis of nitric oxide.3.Zheng's No.1 New Wound Medicine can inhibit the expression of MGF;it can up-regulate the expression of eNOS in a small range,and inhibit the abnormal increase of eNOS,so as to maintain a stable level.
