| Purpose:The application of Traditional Chinese Medicine "Danhuangsan" external to the ulcer wound in diabetic rats,to observe the healing promoting effect on wound tissue and explore the mechanism of diabetic ulcer healing in rats and the role of link.Method:This study feed 36 weight 180-200g,SPF,SD male rats with high fat diet for one month after intraperitoneal injection of streptozotocin induced by 40mg/Kg,to establish the model of diabetic rats(reference standard[1]:third days,seventh days after injection and observe the blood glucose,more than 16.7mmol/L the relative stability of the rats);All the rats were randomly divided into three groups:Traditional Chinese Medicine group,Western Medicine group and the control group,12 rats in each group,the others 12 normal rats were set as normal group;The rats were shaved at the back is flat,with gentian violet marking about 1.5*1.5 the size of the skin,under sterile conditions,cut in the marked area full-thickness skin deep fascia.The Chinese Medicine group was treated by "Danhuangsan" application,the western medicine group were given "growth factor ge",the control group and normal group rats without intervention.Record each rat drug 7 days,14 days,21 days,28 days wound healing rate and the final healing days;Use HE staining,Masson staining and electron microscope to observe the histological changes;For 14 days,28 days of rats were detected by AGEs in serum and growth factor content;Detection of the content of growth factor in neonatal rats with q-PCR.The final healing days and healing rate in 20th day should be recorded.SPSS20.0 software was used for statistical analysis of the number of days of healing,the results of detection of serum markers,and the detection results of tissues.Result:1.Wound healing rate:Chinese Medicine group,Western Medicine group and the control group compared with the control group and normal group,rats in different time after drug intervention point ulcer wound healing rate increased significantly(P<0.05),the Chinese Medicine group is more significantly(P<0.01),four groups of rats with different time points healing rate has the same trend,can be arranged as follows:Chinese Medicine group>the Western Medicine group>normal group>control group;2.The final healing days:Chinese Medicine,Western Medicine group compared with the control group and normal group,rat ulcer wound healing days after drug intervention significantly shortened(P<0.05),the Chinese Medicine group is more significantly(P<0.01),four groups of rats ultimately healing days can be arranged as follows:Chinese medicine<Western Medicine group<normal group<control group;3.the morphological differences of cells:compared with the blank group,the number of new capillaries and fibroblasts in granulation tissue was increased in the traditional Chinese medicine group and Western medicine group;4.biochemical indicators:comparion of Chinese Medicine group,Western medicine group and control group,AGEs in serum of rats decreased after drug intervention;Chinese medicine group and Western medicine group compared with the control group,drug intervention after EGF,VEGF and bFGF level in serum of rats can be improved,and the traditional Chinese Medicine group was significant(P<0.01);comparion of Chinese Medicine group,Western medicine group and blank group,EGF,VEGF and bFGF levels in neonatal rats after drug intervention in the organization can improve,and the traditional Chinese medicine group was significant(P<0.01).Conclusion:1.The application of Chinese medicine "Danhuangsan" is used in the treatment of diabetic rat ulcer can obviously increase the rate of wound healing,shorten the wound healing time,promote the rats diabetes ulcer healing wound healing in rats;2."Danhuangsan" may reduce the level of AGEs in serum of rats,improve the growth factor content in rat serum,promote neovascularization and fibroblast proliferation related;3.The experimental results showed that AGEs was significantly negatively correlated with growth factor,which could promote the healing of diabetic foot wound by reducing the content of AGEs in serum and promoting the growth factor. |
