Objective:To study antibiotic dysbacteria diarrhea and the effect of Qiweibaizhu Powder on the expression of lactase gene in mice,and to explore the correlation between the expression of lactase gene and the effect of intestinal lactase activity in mice,so as to provide basis for the therapeutic mechanism of Qiweibaizhu Powder on dysbacteriosis diarrhea.Methods:42 SPF Kunming mice,half male and half female,were randomly divided into normal group(n=16)and model group(n=24).For 5days of continuous modeling,the normal group was given distilled water0.35 m L/ piece/time,twice daily gavage,while the model group mice were given cefradine capsule and gentamicin sulfate mixed antibiotic11.67 m L/kg/ time,twice daily gavage,causing intestinal flora disorder in mice.After successful modeling,8 mice were selected from the normal group and the model group respectively randomly.After cervical dislocation and death,the contents and intestinal mucosa were collected aseptically to detect the intestinal lactase activity and the total amount of lactase mRNA in intestinal bacteria of mice after modeling of dysbacteriosis diarrhea.On the 5th day of modeling,16 mice were randomly divided into the model group and the Qiwei group randomly in the model group.For 4 days of continuous treatment,the normal group and the model group were given distilled water 0.35 m L/ piece/time,twice a day,and the Qiwei group was given Qiweibaizhu powder 0.104g/kg/ time,twice a day.After 4 days of treatment,8 mice of the three groups were killed respectively to detect the intestinal lactase activity and the total amount of intestinal lactase mRNA in mice after Qiweibaizhu powder was used to treat mice with dysbacteriosis diarrhea.Results: 5 days after the model of dysbacteriosis diarrhea was established,compared with the normal group,the lactase activity in model group were significantly decreased,with statistically significant difference(p<0.01).There was no significant difference in mRNA expression between normal group and model group(p<0.05).After 4 days of treatment with qiweibaizhu powder,compared with the normal group,the lactase activity in the model group decreased,and the lactase activity in the contents(p<0.01)and mucosa(p<0.05)of mice in the qiwei group increased,with statistical significance.Compared with the model group,the intestinal contents and mucosal lactase activity of mice in the Qiwei group were significantly increased,and the difference was statistically significant(p<0.01).There was no significant difference in mRNA expression of mouse contents and mucosa among normal group,model group and Qiwei group(p<0.05).Conclusion: Diarrhea caused by dysbacteriosis can reduce lactase activity in intestinal contents and intestinal mucosa of mice and bacteria,and Qiweibaizhu Powder can promote recovery of lactase activity.Modelling of dysbacteriosis diarrhea and treatment with Qiweibaizhu Powder have no effect on the total amount of lactase mRNA in mice intestinal tract.Qiwebaizhu Powder can regulate the lactase activity in the intestinal tract of mice at the protein level and has no effect on the change of the total amount of lactase mRNA in and mucosa between the normal group and the model group(p<0.05).After 4 days of treatment with Qiwei baizhu Powder,there was no significant difference in the expression of bacterial lactase mRNA in intestinal contents and mucosa of mice in normal group,model group and Qiwei group(p<0.05).Conclusion: Diarrhea caused by dysbacteriosis can reduce lactase activity in intestinal contents and intestinal mucosa of mice and bacteria,and Qiweibaizhu Powder can promote recovery of lactase activity in intestinal tract of mice and bacteria.Modelling of dysbacteriosis diarrhea and treatment with
