Background: Hair is of great value and directly affects beauty.At present,the incidence of alopecia is increasing year by year,and there is a lack of effective external medication in clinic.It is of great significance to explore the mechanism of hair follicle and hair growth and to study the external medication for promoting hair follicle and hair growth.Objective: To explore the effects of Chinese herbal medicine pigments on the growth of mouse hair and hair follicle in vitro,and to study its possible mechanism and prospects.METHODS: Rat back was depilated and randomly divided into control group and experimental group.The experimental group was smeared with safflower yellow,curcumin and shikonin at 10ug/ml,30ug/ml respectively.The positive control group was smeared with oleanolic acid and the negative control group with saline.The skin color,hair growth and histological changes of hair follicles were observed.(2)Separate hair follicles,observe the morphological changes of hair follicles under inverted microscope every other day,determine the length of hair follicles,take photographs and record;MTT check the vitality of hair follicle cells.Result: 1.In vivo study on hair growth: 1.Safflower yellow group and curcumin group can shorten the hair follicle from resting period to growing period when the pigment concentration is 10 ug/ml.Safflower yellow and shikonin have no effect on the final hair length;2.Safflower yellow group,curcumin group and shikonin group can shorten the hair follicle from resting period to growing period when the pigment concentration is 30 ug/ml.(3)Histological sections showed that there were more hair follicles in the three groups of mice,30 ug/ml safflower yellow,10 ug/ml curcumin and 30 ug/ml shikonin,and the growth period was prolonged.2.The results of hair follicle culture in vitro: 1.Compared with the negative control group,curcumin at 3ug/ml and 10ug/ml,shikonin at 30ug/ml and safflower yellow at 30ug/ml could significantly promote hair follicle growth(P < 0.05),and curcumin at 10ug/ml had the strongest effect on hair follicle growth(P < 0.05).2.MTT experiment showed that safflower yellow at 30ug/ml,curcumin at 10ug/ml and shikonin at 30ug/ml had the strongest effect on hair follicle growth(P < 0.05).There was a significant difference in cell viability between the negative control group and the negative control group(P < 0.05),and hair follicle growth was consistent.(3)The relative optimum concentration of pigments showed that the growth of hair follicles was not promoted by the mixed groups,and MTT test showed that the cell viability of the mixed groups decreased significantly.Conclusion: Safflower yellow,curcumin and shikonin can promote the growth of hair and hair follicle,and the concentration of the pigments is different.
