Objective:To investigate the effects of Tongguan Pill-containing serum on the expression of TLR2/My D88/NF-KB signaling pathway-related genes in macrophages induced by mixed bacteria.To explore the mechanism of Tongguan Pill in the treatment of tubal inflammatory obstructive infertility.Methods:A rat vaginal obstructive model was established by vaginal inoculation with mixed bacteria(Staphylococcus aureus,Escherichia coli and Ureaplasma urealyticum).Rats with successful modeling were randomly divided into Tongguan pill group and model group.The Tongguan Pill group was intragastrically administered with a vial suspension,and the model group was given the same amount of normal saline.After 20 days,blood was collected from the abdominal aorta,centrifuged,filtered,and inactivated to prepare serum.The macrophage inflammation model was induced by mixed bacteria,and the drug was treated with Tongguan Pill-containing serum.The cells and supernatant were collected 24 hours later.The expression levels of TLR2,My D88 and NF-κB m RNA were determined by RT-PCR.Results:Compared with the normal control group,the expression levels of TLR2,My D88 and NF-κB m RNA in the mixed bacteria group increased significantly,with significant difference(P<0.01).Compared with the mixed bacteria group,the NF-κB blocker group The expressions of TLR2,My D88 and NF-κB m RNA in the sputum containing Tongguan Pills showed a downward trend,which was statistically significant(P<0.05),but the NF-κB blocker group and Tongguan pill contained There was no significant difference in the expression of TLR2,My D88 and NF-κB m RNA between the serum group and the control group(P>0.05).Compared with the drug-free serum group,the TLR2,My D88 and NF-κB m RNA were contained in the drugcontaining serum group.The expression levels showed a downward trend with significant differences,which was statistically significant(P<0.05).Conclusion:1.A biomodeling method inoculated with a mixed intravaginal bacterium can successfully replicate the model of tubal inflammatory obstructive infertility.2.The regulation of NF-κB activity by regulating the expression of the key elements of the signaling pathway "TLR2,My D88,NF-κB" may be one of the mechanisms of Tongguan Pill in the treatment of inflamed obstructive infertility.
