| Objective:To probe the functional mechanism of the Tongguan pill and to provide an experimental basis for the clinical treatment by studying the effect on the changes of the tubal histomorphology and content of TRL2 and TRL4 in epithelial cells of the rats in SOI model.Methods: 84 SPF Wistar healthy unpregnant rats were used to make ISO Model. According to random number table, rats were divided into 3 groups: normal group(12 r ats), sham-operated group(12 rats) and model group(60 rats). The SOI rat model was m ade by injecting the mixed bacteria. After rat model established, according to random n umber table, the rest rats of the model group were divided into 5 groups: Tongguan pill group(including high-dosage group, medium-dosage group and low-dosage group), mo del controlled group and Fuke Qianjin capsule group, 10 rats in each group. Each grou p were poured with relative medicines into stomach for 20 days before sacrifice. The c hanges of the tubal histomorphology of each group were observed by the HE staining method and the differences among all groups in content of TRL2 and TRL4 in epithelia l cells by immunohistochemistry method.Results: 1.Model evaluation was done respectively in 10 th and 15 th day. The resul t showed acute inflammation in 10 th day and chronic inflammation in 15 th day, which meant the success of model replication. 2.Compared with the model group, the groups by Tongguan pill and Fuke Qianjin capsule group can improve the tubal histomorphology of rats. 3. About the content of TLR2 and TLR4 in epithelial cells, the model group had more positive expression than all other groups, the difference among which were st atistically significant(P<0.05). The postive expression of the high-dosage group was ob viously inferior to it of medium-dosage group, low-dosage group and Fuke Qianjin cap sule group, the difference among which was statistically magnificent(P<0.05). But the difference among medium-dosage group, low-dosage group and Fuke Qianjin capsule group was not statistically significant(P>0.05).Conclusion: 1. The rat SOI model can be established by injecting the mixed bacter ia. 2. The changes of the content of TLR2 and TLR4 are highly related to the occurrenc e and development of chronic tubal inflammatory obstructive infertility. 4. The Tonggu an pill can improve the structure and functions of the rats with tubal inflammation obst ruction infertility and can regulate the content of TLR2 and TLR4 in epithelial cells of t ubal, and therefore has a good effect on treating the infertility with tubal inflammation obstruction. |
