Objective: Through the experiment observe the influence of He shen pills on podocyte skeletal protein synaptopdin in adriamycin-induced nephropathy rats,To investigate the molecular mechanism of He shen Pills in treating proteinuriahe.Methods: 110 healthy and clean-grade male SD rats,urinary protein excretion ?100mg/24 h prompt model copied successfully,The rats were divded into four groups:the normal group(n=25),molde group(n=28),enalapril group(n=28),and Heshen pills group(n=28).Interventing with drugs After the success of the model,8 weeks in all,after 2,4,6 and 8 weeks treatment,rats were killed in batches at four time points,Recording the weight,determined the quantitative of 24-hours urinary protein,ALB,BUN and SCr,Renal tissue was stained with HE and Masson,and observed the pathological structure of kidney under light microscope.The expression of cytoskeleton protein synaptopodin was detected by immunohistochemistry.Results: 1.Heshen pills can improve the general condition of adriamycin nephropathy rats.2.Weight: the weight of the blank group increased faster than that of the other three groups(P<0.05),while the growth of the model group was slower than that of the treatment group(P<0.05).3.Biochemical indexes: compared with the blank group,the quantity of 24 h urine protein,SCr,BUN in the model group were higher than those in the normal group(P<0.05).The treatment group is lower than the model group(P<0.05),Albumin detection,The ALB of model group is lower than the normal group(P<0.05),The level of ALB in the treatment group was higher than that in the model group(P<0.05).There was no significant difference in biochemical indexes between Heshen pills group and enalapril group(P>0.05).4.Pathological observation:The glomerular morphology and structure of rats in blank group were normal.In the model group,the glomerulopathy was not obvious in the early stage of modeling,a little lipid deposition can be seen in renal tubular epithelium,Partial Mesangial cell proliferation,with the increase of time,focal segmental scarring occurs.matrix broadening,vascular loop malopening and collapse,glomerulonephritis and renal vesicle adhesion,Visible protein tube type in collecting tube,tubular vacuolar degeneration,interstitial fibroblast proliferation,with inflammatory cell infiltration,Glomerular and tubulointerstitial lesions in the treatment group werelighter than those in the model group.A small amount of renal interstitial edema,fibrosis,with a small amount of inflammatory cell in filtration.5.immunohistochemical:The expression of synaptopodin protein in the blank group was higher than that in the other three groups(P<0.05).From The fourth week,there was significant difference between the treatment groups and the model group(P<0.05).There was no significant difference between Heshen pills group and enalapril group(P>0.05).Conclusion: The Heshen Pills can reduce urinary protein,Increase serum ALB level,and improve the renal function,Upregulate the expression of cytoskeleton synaptopodin,reduce renal injury.
