Objective: To investigate the expression of MCP-1 in AIN rats urine and renal tissue Observation enalapril on its expression .Study the protective effect of Enalapril on kidney ,the role of MCP-1 in i proteinuria in minimal change nephropathy syndrome (MCNS). Methods: Male Wistar rats 140. Were randomly divided into normal group, nephropathy group. Normal 40 (random further divided into four groups, each with 10). Injection of saline 5 mg / kg; Nephropathy group 100, a single rat tail vein injection of doxorubicin 5 mg / kg; Preparation MCNS model. All the rats of the 24-hour urine protein quantitative detective on the sixth day after injection .urine protein> 30 mg/24h are successful model for MCNS. Removed four.Successful nephropathy rats models (total of 96)randomly divided into two groups again . Nephropathy control group (n = 49), the treatment group (n = 47). Treatment group to Enalapril 2 mg / kg dissolved in 1 ml of physiological saline. In the seventh day after the s modeling administration. Nephropathy control group and the normal group to the contour NS gavage, At the end of the 7,14,28,42 day detective all the groups of the rats 24-hour urine protein and serum albumin and serum total cholesterol levels .At the same time using the sandwich ELISA detective the MCP-1 level of rat urine and kidney . And the rats were sacrificed, Renal tissue from rats doing light microscope examination, investigate the changes of renal morphological. Results: 1. In different periods nephropathy control rat urine and kidney tissue of MCP-1 was significantly higher than the same period in normal group of MCP - 1 value. There were significant differences, and different periods of MCP-1 expression has different degrees. 2. In different periods the value of rats urine and kidney tissue of MCP-1 lower than the control group nephropathy low, but higher than the normal group, there were significant differences. 3. In different periods in urine and kidney tissue of MCP-1 the changes in 24-hour urine protein was positively correlated. MCP-1 in urine and kidney concentration of MCP-1 levels are positively correlated. Conclusion: 1. Adriamycin single intravenous injection of 5 mg / kg MCNS model can be successfully prepared. 2. Adriamycin nephropathy in rats urine MCP-1 in renal concentration of MCP-1 were significantly increased, and with 24-hour urine protein levels were positively correlated, suggesting that MCP-1 is involved in a large doxorubicin nephropathy Rat proteinuria occurred. 3. Both the levels of MCP-1 in urine and kidney were positively correlated, the level of MCP-1 in urine can reflect the MCP-1 level in renal tissue .levels of MCP-1 in urine can be used as detection indicators in MCNS disease 4. Enalapril can be by influencing the expression of MCP-1 to reduce proteinuria, these drugs may be one of the mechanisms to protect the kidney.
