ObjectiveChoosing spontaneous hypertensive rats(SHR)as experimental object,we can observ the intervention effect of electroacupuncture stimulation of "Taichong"(LR 3)and "Baihui"(GV 20)on spontaneously hypertensive renal damage through the comparison between groups of rats systolic blood pressure,serum uric acid(UA)and the content of urea nitrogen(BUN)and light microscopy,electron microscopy,the renal pathological structure changes.In addition,through the analysis of each group in the rat kidney angiotensin-converting enzyme 2(ACE2),angiotensin 1-7(Ang(1-7),angiotensin ?(Ang ?)expression,the aim is to further explore the possible mechanisms of renal protection in spontaneously hypertensive rats.Methods24 SHRs were randomized into model and Electroacupuncture(EA)group with 12 Wistar-Kyoto(WKY)rats as normal group.The EA group received electroacupuncture at right side of Taichong ang Baihui(1mA,2Hz/15Hz,for 20min),once per day for 30 days;The other groups were given the same grasping and fixation stimulation as the electroacupuncture group except the electroacupuncture.The systolic blood pressure(SBP)variation condition of rats in each group was monitored adopting noninvasive blood pressure instrument before intervention and in the 6nd,the 12th,the 18th,the 24th and the 30th day during intervention.After 30 days treatment,the changes of blood uric acid(UA)?blood urea nitaogen(BUN)?ACE2?Ang(1-7)?Ang? and Ang(1-7)/Ang? was determined.Pathological changes of renal tissues in rats were observed by HE?Masson staining and Electron microscope.Results1.Observation of systolic blood pressure of rats before and after treatment in each groupThe systolic blood pressure of the model group and electroacupuncture group was significantly higher compaired the blank group before treatment,and the difference was statistically significant(P<0.01),However,there was no difference in systolic blood pressure between the model group and the electroacupuncture group(P>0.05).After 24 days of treatment,the systolic blood pressure of the electroacupuncture group was lower than that of the model group,and the difference was statistically significant(P<0.05).After 30 days of treatment,there were significant statistical differences between the electroacupuncture group,the blank group and the model group(P<0.01),On the whole,The overall trend is as follows:Model group including systolic blood pressure>systolic blood pressure>blank systolic blood pressure.2.Analysis of serum uric acid(UA)and urea nitrogen(BUN)content in each group Compared with the blank group,the contents of UA and BUN in the serum of the model group were significantly increased(P<0.01).After 30 days of electroacupuncture treatment,the contents of UA and BUN in the serum of the electroacupuncture group and the model group were decreased,and the results were statistically significant(P<0.05).3 Renal tissue converting enzyme 2(ACE2),angiotensin 1-7(Ang(1-7),angiotensin ?(Ang?)expression analysis in three groups Model group compared with the blank group,kidney ACE2,Ang(1-7),Ang(1-7)/Ang ?expression were significantly lower,Ang ? expression increased significantly,differences are statistically significant differences(P<0.01);After a cupping treatment,the electroacupuncture group compared with model group,kidney ACE2 expression increased,the difference was statistically significant(P<0.05)and Ang(1-7),Ang(1-7)/Ang ? expression increased significantly,with significant difference statistically significant(P<0.01),Ang? expression decreased,the difference was statistically significant(P<0.05);There was no significant difference in ACE2 expression between the electroacupuncture group and the blank group(P>0.05).4.Observation of renal histology in each groupThe results of HE staining showed that compared with the blank group,the model group had significantly dilated glomerular cystic space,irregular arrangement of renal tubules,and vacuolar degeneration of renal tubular epithelial cells.Compared with the model group,the electroacupuncture group showed less renal damage.Masson staining showed that the degree of collagen accumulation in the electroacupuncture group was significantly lower than that in the model group.The results of electron microscopy showed that in the model group,the morphology and structure of the pedocytes were irregular,the fusion was obvious,the mitochondria were swollen,and mitochondrial vacuoles appeared.Compared with the model group,the morphology of the foot processes was relatively regular,the degree of fusion was reduced,and the mitochondrial swelling was not obvious.ConclusionElectroacupuncture at Taichong(LR?3)and Baihui(GV?20)could effectively reduce systolic blood pressure(SBP)and the damage to the kidney,The mechanism may be through increased kidney ACE2?and Ang(1-7)expression,at the same time suppress Ang ? protein expression,increase Ang(1-7)/Ang ? ratio.
