Objective:Based on MAPK signaling pathway,the differences of p-JNK?P38and TG between model control group,treatment group and blank group were observed.And through more subtle and objective observation of the changes in rabbit femoral head bone tissue specimens,to study the mechanism of action of Chuangu tablets in the treatment of SANFH,to offer a new means for it.Methods:Two groups were randomly separated from the 40 male rabbits of Japanese: the blank group and the model group.After 1 weeks of feeding,the model was established by the method of "He Xi Jing",with a period of 6 weeks.After 6 weeks,4 rabbits were chosen randomly in each group to identify whether the mold is successful.After modeling,the model groups were randomly divided into model control group,Chuangu tablet treatment group and Xianlinggubao capsule treatment group,with 8rabbits in each group.The latter experiment was converted according to the 60 kg body weight conversion algorithm of adults.The treatment group was given gavage with medicine every day.The model control group was given distilled water every day,while the blank group was given no treatment for 42 days.After 42 days,collecting the abdominal aortic blood to calculate the serum TG concentration of each group,and remove the right femoral head pathological sections.The tissue morphology of bone tissue was observed,and p-jnk and p38 protein contents were detected.Results:1?p-JNK:After 13 weeks,contrasted with the model matched group,the p-jnk gene expression in the treatment group was significantly down-regulated(p=0.000,p=0.000),while the comparison between the Chuangu tablet group and Xianlinggubao capsule group showed no statistical significance(p= 0.535).2?P38:After 13 weeks,contrasted with the model matched group,the P38 gene expression in the treatment group was significantly down-regulated(P<0.05),which was significant,while the difference between Chuangu tablet group and Xianlinggubao capsule group has no statistically significant(P>0.05).3?TG: After 13 weeks,compared with the model control group,the serum TG content of the treatment group was reduced(P < 0.05),with statistical significance.However,there was no significant difference between Chuangu tablet group and Xianlinggubao capsule group(P>0.05).Conclusion:1?Chuangu tablet may improve the cytothesis of bone cells,and suppress the apoptosis rate of cells,which is negatively correlated with the expression of p-jnk and p38 protein.Its relationship with p-jnk and p38 protein genes is still unclear,and the mechanism of interaction between p-JNK and p38 pathways needs to be further explored.2?Chuangu tablet may down-regulate the expression of p-jnk and p38 protein genes,slow down the apoptosis of bone cells,reduce the incidence of empty bone lacunae in femoral head,and play a role in the early treatment of steroid-induced femoral head necrosis.3?Chuangu tablet may promote the differentiation of bone stromal cells(BMSCs)into osteoblasts,thus playing an important role in the repair of necrotic bone cells.
