The Effect Of Wenjing Decoction On The Behavior And Amino Acid Neurotransmitters In The Mouse Model Of Tic Disorder

Purpose:To observe and discuss the effectiveness and mechanism of Wenjing Decoction(WJD)in TD mouse model?Material and method:Seventy-two male ICR mice were randomly divided into normal group,model group,western medicine group(Tiapride group),WJD high,middle and low dose group,each group of 12.IDPN intraperitoneal injection of 450mg/(kg·d)10d induced TD mouse model.After successful model to medicine,normal group and model group to 0.9% sodium chloride solution,3 groups of WJD to the traditional Chinese medicine 56 g,28g and 14 g/(kg·d)respectively,western medicine group in Tiapride will be the 25 mg/(kg·d)for lavage treatment.once a day,and the grading of routine and motor behavior was determined after 28 days of continuous administration.HE was used to observe the pathological changes of each group of mice and choose the ELISA method detect contents of GLU,ASP and GABA in the striatum of mice?Results: 1.Behavioral testing:The behavioral scores of the mice in each group were significantly lower than that in the model group(P < 0.05)?2.HE staining result:Quiet decoction high dose group and western medicine group closest to the result of the normal group,200 times under the microscope observation,visible mouse cerebral cortex,striatum were better recovery,the morphology,structure neurons long axons and dendrites,short,thick,thin improved,arrange the rules,infiltration of inflammatory cells and necrosis of neurons compared with model group.?3.Determination of GLU,ASP and GABA in brain tissue:Compared with normal group,model group,GLU,ASP content of brain tissue increased significantly(P < 0.01),compared with model group,the rest of the group have a trend of decrease,WJD middle and high dose group and western medicine group gentle difference(P>0.05).compared with normal group GABA levels,model group were decreased significantly(P<0.01),compared with model group,the rest of the group are a trend of increase,WJD high dose group and western medicine group have gentle difference(P>0.05)Conclusion: 1.The experiment showed that IDPN could induce behavioral symptoms similar to TD in mice.?2.WJD can inhibit and alleviate the stereotyping and motor behavior of TD mice?3.Its effect may be associated with the inhibitory effect of GLU,ASP content and stimulation of GABA,and the high dose group in WJD is comparable to that of the western medicine group?...
