Purpose: Through observing the effect of LBP on the metabolism of the lipid in serum and the expression of protein and m RNA in HMGCR,L-FABP,LPL in livers of the ovariectomized rats,to probe into the mechanism of LBP on lipid metabolism,Therefore,it may provide an experimental basis for the prevention and treatment of coronary heart disease caused by abnormal lipid metabolism in perimenopausal women.Material and method: 30 SPF healthy female SD rats,adaptive feeding and drinking,were randomly divided into 5 groups after the operations,6 of them,kept the ovaries and only removed a small amount of adipose tissue around the uterus,were treated as blank control group or sham operation group.The remaining 4 groups had bilateral ovaries removed and respectively were ovariectomized group,Progynova group,LBP high dose group and LBP low dose group.After the operations,the muscle was injected with penicillin for 1 week to keep the rats free from infection.The sham operation group and the ovariectomized group were treated by gavage with the same amount of physiological saline.The rats in Progynova group were treated by gavage with Estradiol valerate.LBP high dose group and LBP low dose group were treated by gavage with different doses of LBP.The rats treated for 12 weeks and the levels of TC,TG,LDL-C,HDL-C in serum were detected out by automatic biochemical analyser.Using the HE staining to observe the morphological changes of liver tissues in rats.Detecting out the HMGCR,L-FABP and LPL in livers by the western blot.Detecting out the expression of HMGCR m RNA,L-FABP m RNA and LPL m RNA in livers by the RT-RCR technique.Results: 1.Serum lipids detection found that compared with the sham operation group,TC and LDL-C were significantly increased and HDL-C was decreased significantly in ovariectomized group(P<0.01);Compared with ovariectomized group,the level of TC in Progynova group andLBP high dose group were decreased significantly(P<0.01),the levels of LDL-C in Progynova group,LBP high dose group and LBP low dose group were decreased significantly(P<0.01 or P<0.05).2.HE staining found that compared with the sham operation group,there were fat cavitations in ovariectomized group livers,while fat cavitations in Progynova group and LBP high dose group were significantly decreased.The fat cavitations were still visible in the LBP low dose group.3.According to the results of western blot,compared with the sham operation group,the expression of HMGCR and L-FABP protein were increased significantly(P<0.01)and the expression of LPL protein was decreased significantly in ovariectomized group(P<0.01);Compared with ovariectomized group,the expression of HMGCR protein was decreased and the expression of LPL protein was increased significantly in Progynova group(P<0.05 or P<0.01).The expression of HMGCR and L-FABP protein were decreased significantly,the tendency about the expression of LBP was opposite in LBP high dose group(P<0.01).There was no significant difference in the expression of protein in the LBP low dose group.4.According to the results of RT-PCR,compared with the sham operation group,the levels of HMGCR m RNA and L-FABP m RNA were increased significantly(P<0.01),and the level of LPL m RNA was decreased significantly(P<0.01).Compared with the ovariectomized group,the levels of HNGCR m RNA and L-FABP m RNA were decreased,but the level of LPL m RNA was increased significantly in Progynova group(P<0.05 or P<0.01).The level of HMGCR m RNA was decreased,the level of L-FABP m RNA was decreased significantly in and the level of LPL m RNA was increased significantly in LBP high dose group(P<0.05 or P<0.01).There was no significant difference in the expression of m RNA in the LBP low dose group.Conclusion: 1.LBP can improve lipid metabolism disorders effectively of ovariectomized rats.2.LBP may improve the lipid metabolism disorders of ovariectomized rats by means of down-regulation on the protein of HMGCR,L-FABP and up-regulation on the protein of LPL with dose-effect relationship in livers.
