Abstract/Summary:
In recent years,as the incidence of malignant tumors has increased year by year,its damage to human life and health has also received close attention.Among them,the incidence and mortality of breast cancer ranks first in female malignant tumors,making it the number one killer of women’s life and health in malignant tumors.As a huge treasure house left by our ancestors,traditional Chinese medicine uses modern medicine and pharmacology to conduct research,and can further explore new anti-tumor drugs and active substances to prevent and treat malignant tumors.At the same time,when a variety of traditional Chinese medicines are used in combination with commonly used chemotherapeutic drugs,they can have effects such as increasing efficacy,reducing toxicity,and preventing drug resistance.Therefore,the development of traditional drugs or active substances is also of great significance for the treatment of tumors.In this study,the active ingredient baicalein in Astragalus membranaceus was the main research object.Although it has many physiological activities,its mechanism of action on human breast cancer is not completely clear.Therefore,this experiment combined the anti-tumor study of baicalein and the first-line drug docetaxel in breast cancer to explore the mechanism of safrole inhibiting tumor cell proliferation.The results of this study will be the development and utilization of Chinese herbal medicine Astragalus and its effective active ingredients.Provide basic theoretical basis.Purpose:Based on human breast cancer MCF-7 cell line,the combination of docetaxel,baicalein,docetaxel and baicalein was used to study the biological activity of drug-treated cell lines,and further study on different drugs The mechanism of tumor function.Method:1.The human breast cancer MCF-7 cell line was used as the experimental study.CCK-8method was used to determine the inhibition rate and cell viability of cells treated with different concentrations of docetaxel and baicalein at specific time.According to the experimental data,the inhibition rate and cell viability of docetaxel + baicalein combined treatment of human breast cancer MCF-7 cell line were determined.The synergistic effect was calculated according to the formula.2.According to the CCK-8 results,select the appropriate drug concentration and treatment duration.The apoptosis rate of human breast cancer MCF-7 cell line after drug treatment was detected by flow cytometry using Annexin V-PI double staining.The apoptosis cycle ofhuman breast cancer MCF-7 cell line after drug treatment was detected by flow cytometry using PI(propidium iodide)single staining.3.Western blot was used to detect the expression levels of MAPK/JNK pathway-related proteins JNK,p-JNK,p53,Caspase-3 and Caspase-9,thereby exploring the anti-tumor effect of baicalein,docetaxel and baicalein.Ways and mechanisms.Result:1.Docetaxel alone and baicalein inhibited the growth of human breast cancer MCF-7 cell line in a time-dose dependent manner.Different concentrations of baicalein combined with docetaxel inhibited the growth of human breast cancer MCF-7 cell line in a time-dose-dependent manner,and baicalein enhanced docetaxel on human breast cancer MCF The proliferation inhibition effect of the-7 cell strain has an additive or synergistic effect depending on the concentration.2.Flow cytometry results indicated that baicalein can significantly enhance the apoptosis of human breast cancer MCF-7 cell line induced by docetaxel.The mechanism of apoptosis may be related to docetaxel blockade of human breast cancer MCF-7.The cell line was arrested in the G2/M phase,the baicalein monotherapy group and the docetaxel + baicalein group in the G0/G1 phase.3.Western blot analysis showed that docetaxel alone could promote the expression of P53 gene,increase the cleavage of Caspase-3 and Caspase-9 protein,increase the expression of JNK protein,but express the phosphorylated p-JNK protein.no effect.The single drug of baicalein can promote the expression of P53 gene,increase the cleavage of Caspase-3 and Caspase-9 protein,and increase the expression level of p-JNK protein.The combination of docetaxel and baicalein significantly increased the cleavage of Caspase-3 protein and increased the expression level of p-JNK protein.It indicated that the combination of docetaxel and baicalein could promote the expression of P53 gene and increase the cleavage activation of Caspase-3 and Caspase-9 by MAPK/JNK pathway,and finally enhance the docetaxel-promoting breast cancer MCF.The role of-7 cell line apoptosis.Conclusion:The data suggest that baicalein can inhibit the growth of human breast cancer MCF-7 cell line and induce its apoptosis.The mechanism may be to promote the expression of P53 gene and activate Caspase family Caspase by regulating MAPK/JNK signaling pathway Caspase-3 and Caspase-9 protein,arrest cell cycle,etc.,finally induced growth inhibition and cell apoptosis of human breast cancer MCF-7 cell line.At the same time,the combination of baicalein anddocetaxel has a synergistic effect on tumors,which may also be related to the MAPK/JNK signaling pathway.
