Purpose: This study was to explore the mechanism of the effect of Shenfukang ? in the treatment of adriamycin-induced nephropathy in rats with adriamycin-induced nephropathy,and to study the effect of Shenfukang ? on the glomerular molecular barrier.In order to provide a new evidence for the treatment of nephrotic syndrome in the clinic,which is characterized by the micro-pathological changes,it provides a theoretical basis for clinical application of the kidney-Fukang ?.Materials and Methods: 60 healthy male SD rats were randomly divided into the normal group,the model group,the traditional Chinese medicine group,the hormone group and the combination group.In addition to the normal group,the other groups were injected with the tail vein for 2 times,and the model of the adriamycin-induced nephropathy rats was established.After the modeling,the groups were given corresponding drug intervention,and the drug was administered for 4 weeks.The serum albumin,blood lipid and kidney function were measured at the fourth weekend after the first week and the fourth week of treatment,and the pathological changes of the kidney of the rat were observed under light microscope.The expression of CD2 AP,nephrin and podoxin was determined by immunohistochemistry.The single-factor analysis of variance among the groups was performed with SPSS 20.0software,and the data was expressed by xundefineds.The comparison between the groups was the LSD-t test.Results:1.The model was established: after 2 weeks,the 24-hour urine protein quantitative model group was higher than that of the normal group(P <0.05),and each drug intervention group was compared with the model group(P> 0.05),and the model was suggested to be successful.2.General status: after modeling,except for the other groups of the normal group,the color difference and the loss of the hair,the control of the acousto-optic stimulation response to the normal group was significantly reduced,the amount of intake water was reduced,the stool was lean,and the urine volume was less than that of the normal group,and the scrotum,thepaw edema and the ascites were found in all the rats except the normal group.The general status of each treatment group was improved after administration,and the amount of urine in the western medicine group and the combination group was higher than that of the model group(P <0.05).3.24 h the quantitative change of urinary protein: in addition to the normal group,the quantitative determination of 24 h urinary protein in the other group was higher than that in the normal group(P <0.05).In the first week of administration,the normal group was lower than the other group(P <0.05),and there was no statistical difference between the model group and the treatment group(P> 0.05).In the same period,the mean value of urinary protein in the model group was higher than that of the other treatment group(P <0.05),and the mean value of the combination group was lower than that of the other treatment group(P<0.05),and the western medicine group and the traditional Chinese medicine group had no statistical significance(P> 0.05).4.The change of biochemical index: the serum albumin,the normal group was superior to the model group and the drug intervention group(P <0.05),the drug intervention group was superior to the model group(P <0.05),and the drug intervention group had no statistical significance(P> 0.05);The normal group of the serum triglyceride was lower than the other groups(P <0.05).Only the western medicine group and the combination group were lower than the model group(P <0.05)in the drug intervention group,and there was no statistical significance between the treatment groups(P> 0.05).There was no significant difference between the treatment groups and the model group(P> 0.05),and there was no significant difference between the treatment groups(P> 0.05).There was no statistical significance between the other groups except that the model group was higher than that of the normal group(P <0.05).There was no significant difference between the relevant values of urea and nitrogen(P> 0.05).5.In the aspects of the pathological change of the kidney,the mesangial cells of the model group were seen to be hyperplastic,and the mesangial cells of the drug-intervention group had no obvious proliferation,and the tubular protein-type and the degree of renal interstitial edema were reduced in the traditional Chinese medicine group,and the hormone group and the combination group were reduced.There was no significant difference between thecombination group and the hormone group.6.The expression of nephrin,nephrin and CD2 AP was observed in the renal tissues of each group,and the average optical density of nephrin was lower than that of the normal group(P<0.05).In addition to the western medicine group,the expression of the other drug intervention group was higher than that of the model group(P <0.05),and the comparison between the traditional Chinese medicine group and the combination group was not statistically significant(P> 0.05),and the average optical density of the renal tissue podoxin was lower than that of the normal group(P <0.05).In the model group,only the combined group expression was higher than that of the model group(P <0.05),and the average optical density of CD2 AP was not statistically significant(P> 0.05).Conclusion:1.The model of adriamycin-induced rat was successfully produced in this experiment.2.Shenfukang ? can improve the general status of adriamycin-induced nephropathy rats,reduce the content of 24-hour urine protein in adriamycin-induced nephropathy rats,and reduce the content of serum triglyceride,and the serum cholesterol,The effect of serum albumin intervention is not obvious,and the renal pathological state of the adriamycin-induced nephropathy rats can be reduced or relieved,and the combined hormone prednisone therapy can partially enhance the intervention effect.3.The effect of Shenfukang ? in the treatment of adriamycin-induced nephropathy may be related to the improvement of the expression of podoxin,nephrin and the partial repair of the molecular barrier of adriamycin-induced nephropathy rats.
