The Effect Of Baihujia Ginseng Decoction On The Intestinal Mucosal Barrier Of MKR Transgenic Type 2 Diabetic Mice

Objective:Through sorting out and summarized the relevant provisions of zhang zhongjing's theory of spleen and stomach and theory of Xiaoke,and deeply discussed zhang zhongjing's rule of treating quenching thirst based on syndrome differentiation with spleen and stomach as the center.MKR mice were used as experimental subjects,and Baihu and Renshen decoction were used as representative prescription for animal experiments,guided by zhang zhongjing's theory of treating Xiaoke from the perspective of spleen and stomach.To study the scientific connotation of Xiaoke from spleen and stomach of zhang zhongjing by observing the effects of Baihu and Renshen decoction on blood glucose and intestinal barrier of MKR mice.Methods: In the literature part,Sorting out the literature context of the ancient concept of Xiaoke by means of literature research to determine the meaning of related concepts of "Xiaoke" and analyze the value of the thought of treating Xiaoke with spleen and stomach to modern understanding and treatment of diabetes.In the experimental part,a total of 32 MKR mouse was given STZ intraperitoneal injection after after 2 weeks on the high fat emulsion.Then they were randomly divided into model group,low dose Baihu and Renshen decoction group(13.5g/kg),high dose Baihu and Renshen decoction group(54g/kg)and metformin group(200 mg/kg).the mice were sacrificed after 28 days intragastric administration,fasting blood glucose(FBG)was determined by electrochemical method,the levels of interleukin-6(IL-6),interleukin-1?(IL-1?),tumor necrosis factor-?(TNF-?)and lipopolysaccharide(LPS)in serum were examined by ELISA.The morphological changes of colon tissue were observed by hematoxylin-eosin(HE)staining.The protein expression of ZO-1?Claudin-1?Occludin in colon were measured by Western blot.Results:The results of literature research show that zhang zhongjing believes that the spleen and stomach can transport water and water,and that the spleen and stomach are the hub of human qi and the source of qi and blood.It is believed that the pathogenesis and treatment of internal injury and miscellaneous diseases are related to spleen and stomach,and it is suggested that the pathogenesis of relieving Xiaoke lies in spleen and stomach.The results of Animal studies shown that compared with model group,FBG were significantly decreased(P<0.01)and serum IL-6,IL-1?,TNF-?,LPS were decreased(P<0.01,P<0.05)in each administration group.The mucosal epithelium integrity improved.The protein expression levels of Claudin-1,Occludin were increased(P<0.01,P<0.05)in high dose Baihu and Renshen decoction and metformin group,while ZO-1protein expressions showed no statistical difference in high dose Baihu and Renshen decoction group.And ZO-1,Claudin-1,Occludin protein expressions showed no statistical difference in low dose Baihu and Renshen decoction group.Conclusion: The fundamental pathogenesis of Xiaoke was Spleen deficiency and stomach heat,and invigorating qi,nourishing jin and clearing lung and stomach represented by Baihu and Renshen decoction were the important methods of treating Xiaoke.Baihu and Renshen decoction could consequently reduce the levels of LPS and proinflammatory cytokines in MKR mice and increased the protein expression levels of tight junction protein in intestinal tract.Baihu and Renshen decoction could lower FBG and control T2 DM by amelioratingintestinal mucosal barrier and attenuating endotoxaemia and inflammation.
