| Purpose:To observe the effect of moxibustion on the total protein content in the skeletal muscle of rats with gastric cancer,and to explore the possible mechanism of moxibustion reducing the protein consumption of skeletal muscle of rats with gastric cancer,from the effect of NF-κ b activation on the expression of the rate limiting enzymes Mu RF1 and MAFbx m RNA in the ubiquitin protease system,the main pathway of muscle protein degradation.Methods:43 male healthy SPF grade SD rats were divided into model group(10 rats)and moxibustion group(10 rats).Twenty rats without gastric cancer model were randomly divided into two groups: blank group and sham operation group.After grouping,moxibustion group was given alternate mild moxibustion every other day:(1)Zhongwan,Guanyuan,Zusanli,(2)Pishu and Weishu.The blank group,the sham operation group and the model group were accompanied by grasping and binding fixation.Each group was intervened for 20 minutes once a day for 14 days.At the end of the treatment,the animals were killed,the cancer weight and cancer volume were measured,the tumor inhibition rate was calculated,the body mass of the organs removed and the weight of the gastrocnemius muscles were measured,Western blot was used,WB)method was used to detect the total protein content in gastrocnemius.The levels of NF-Κb,IΚb-α and PIΚb-αin skeletal muscle were detected by WB method and immunohistochemistry respectively.The expression of Mu RF1 and MAFbx m RNA in skeletal muscle was detected by fluorescence quantitative PCR.Result:1.After the intervention,compared with the model group,the cancer weight and tumor volume of moxibustion group decreased(P<0.01),and the tumor inhibition rate was 54.78%.2.After the intervention,compared with the blank group,the body weight of the rats in the sham operation group had no difference(P > 0.05);compared with the blank group and the sham operation group,the body weight of the rats in the model groupdecreased significantly(P < 0.01);compared with the model group,the body weight of the rats in the moxibustion group increased significantly(P < 0.01).3.After the intervention,compared with the blank group,the quality of gastrocnemius muscle in the sham operation group had no difference(P > 0.05);compared with the blank group and the sham operation group,the quality of gastrocnemius muscle in the model group decreased significantly(P < 0.01);compared with the model group,the quality of gastrocnemius muscle in the moxibustion group increased significantly(P <0.01).4.After the intervention,compared with the blank group,there was no difference in total protein in gastrocnemius muscle of rats in the sham operation group(P > 0.05);compared with the blank group and sham operation group,the content of gastrocnemius muscle protein in the model group decreased significantly(P < 0.01);compared with the model group,the content of gastrocnemius muscle protein in the moxibustion group increased significantly(P < 0.05).5.After the intervention,compared with the blank group,there was no difference in Mu RF1 and MAFbx m RNA expression in the gastrocnemius muscle of the sham operated group(P>0.05);compared with the blank group,Mu RF1 and MAFbx m RNA expression in the gastrocnemius muscle of the model group increased(P<0.05);compared with the sham operated group,Mu RF1 and MAFbx m RNA expression in the gastrocnemius muscle of the model group was increased(P<0.05)Compared with the model group,the expression of Mu RF1 m RNA in the gastrocnemius muscle of moxibustion group decreased(P<0.05),and the expression of MAFbx m RNA decreased significantly(P<0.01).6.After the intervention,compared with the blank group,there was no difference in NF-Κb,PIΚb-α and IΚb-α in the gastrocnemius muscle of rats in the sham operation group(P>0.05);compared with the sham operation group,NF-Κb in the gastrocnemius muscle of rats in the model group increased significantly(P<0.01),PIΚb-α decreased significantly(P<0.01),IΚb-α decreased significantly(P<0.01);compared with the model group,NF-Κb protein in the gastrocnemiusmuscle of rats in the moxibustion group contained significantly The contents of IΚb-αprotein and PIΚb-α protein decreased significantly(P<0.05).Conclusion:1.Moxibustion can inhibit the growth of tumor weight and volume in gastric cancer rats.2.Moxibustion can inhibit the body weight and skeletal muscle consumption of gastric cancer rats.3.Moxibustion can significantly increase the total protein content in gastrocnemius muscle of rats with gastric cancer.Its effect may be related to reducing the activation of IΚb-αand NF-Κb,the expression of Mu RF1 and MAFbx m RNA,and the degradation of protein by ubiquitin proteasome pathway. |
