Objective: The aim of this study is to evaluate clinic Value whether thecombined Survivin mRNA with cytokeratin19(CKl9)mRNA would improvethe diagonosis of gastric cancer and identify early micrometastases status bydetecting the expression of Survivin mRNA and cytokeratin19(CKl9)mRNAin peripheral blood of gastric cancer patients.Methods:We used reverse transcription polymerase chain reaction(RT-PCR)to determine the expression of Survivin mRNA and CK19mRNA inperipheral blood of50gastric cancer patients,30of benign gastric diseasesand30of healthy persons as control, and analyzed the relationship of theexpression and clinical pathological factors. The data was analyzed by x2text.Results:(1)The level of Survivin mRNA and CK19mRNA in peripheralblood of gastric cancer group is higher than normal group and benign gastricdiseases group.(2)The level of Survivin mRNA and CK19mRNA in gastriccancer patients’peripheral blood is correlated with the TNM staging,but notcorrelation with the sex,age,size of tumor and degrees of differentiation.(3)Sensitivity of detection Survivin mRNA combined with CKl9mRNA washigher than that of separate detection.Conclusion:(1) RT-PCR could be a sensitive and specific method toexamine the micrometastases in peripheral blood of patients with gastriccancer.(2) Micrometastases probably presents in peripheral blood of patients with early gastric cancer.(3) The level of Survivin mRNA and CK19mRNAin plasma of patients has some correlation with tumor’s invasion, metastasisand relapse.(4) To detect Survivin mRNA and CK19mRNA in plasma,especially to joint detect Survivin mRNA and CK19mRNA in plasma hasimportant clinical significance in treatment, diagnosis and prognosis.
