Objective:At present,5-fluorouracil(5-FU)has been clinically found to be the main chemotherapeutic agent in the treatment of colon cancer,and its decreased sensitivity to chemotherapy often leads to poor treatment results.However,studies have found that the antitumor effect of curcumin,a traditional Chinese medicine,can increase the sensitivity of chemotherapeutic drugs,thereby inhibiting epithelial-mesenchymal transition(EMT).Therefore,this study explores the sensitization effect of curcumin combined with 5-FU on colon cancer cells based on cell proliferation,apoptosis,migration and metastasis,and provides theoretical and experimental basis for clinical treatment of colon cancer.Methods:This experiment used curcumin and 5-FU alone and in combination to induce colon cancer cell apoptosis and inhibit invasion and metastasis.MTT method and clone formation test were used to detect cell proliferation;Compusyn software was used to analyze the effect of curcumin on 5-FU;scratch test was used to detect cell migration ability;AnnexinV-FITC flow cytometry was used to detect the effect of drugs on colon cancer cell apoptosis;Western blot analysis of intracellular apoptosis-related proteins and N-cadherin,E-cadherin,snail,?-catenin and vimentin protein expression levels.Results:Both curcumin and 5-FU can inhibit the growth of colon cancer cells,but 5-FU lacks sensitivity to SW620 and HCT8 cells.Curcumin exerts a sensitizing effect on 5-FU in colon cancer cell lines,and synergistically inhibits cell proliferation and induces apoptosis.The combined application of the two drugs reduced the migration capacity of colon cancer cell lines,up-regulated the expression of Cleaved caspase-3 and E-Cadherin protein,and inhibited the expression of bcl-2,N-Cadherin,snail,?-catenin,and vimentin.Conclusion:Curcumin exerts a sensitizing effect on 5-FU in colon cancer cell lines,and can synergistically inhibit colon cancer cell proliferation and induce apoptosis,reduce the ability to migrate,and thereby inhibit invasion and metastasis.Its mechanism of inhibiting metastasis may be related to the down-regulation of snail expression by inhibiting the Wnt/?-Catenin signaling pathway,and ultimately inhibiting colon cancer cell EMT.
