Gastric cancer is one of common malignant tumors of the digestive system,its insidious onset,limited comprehensive treatment,the mortality rate remains high,is one of the major clinical cancer.Due to the imperfections of the health care system,health insurance policy is not perfect,food safety problems,the lack of awareness of health care and other issues,leading to emergence of early diagnosis of gastric cancer is low,a high proportion of patients with advanced,high mortality and so on.Current clinical treatment guidelines have proven chemotherapy treatment of gastric cancer is a common means of clinical and surgical conditions and have not applied to tumor recurrence after surgery,the main treatment options metastasis.Chemotherapy(Fluorouracil,5-FU)is currently the main Treatment.It pointed out that a large number of evidence-based medicine,a variety of chemotherapeutic agents in combination with programs such as FOLFOX6 regimen(oxaliplatin,fluorouracil,leucovorin),paclitaxel and5-fluorouracil in combination with chemotherapy programs such effect has been more significant improvement late survival rate has been greatly improved.Since the effective dose and toxic dose chemotherapy too close,will be accompanied by chemotherapy,bone marrow suppression,nausea,vomiting,liver and kidney dysfunction and other side effects of a strong stimulus,more effective,but also increased the patient's pain,greatly reduces the patient post-treatment quality of life,as well as to patients caused great psychological burden.Some patients can not tolerate due to side effects of5-FU and termination of chemotherapy,but also greatly reduces the life quality of patients with late stage.Recent studies show that curcumin have multiple pharmacological functions,such as anti-oxidant,anti-inflammatory,improve immunity and anti-tumor.Wherein the direction of curcumin anti-tumor extensive attention of scholars.Curcumin as an anti-tumor drug or chemotherapy sensitizer is used in a variety of tumor cells and animal models.Objective: This paper aims to study curcumin with the low dose5-fluorouracil(5-FU)on proliferation,apoptosis and cell cycle in gastric cancer MGC-803 cells for the clinical application of curcumin as a gastric cancer chemotherapeutic agent 5-FU synergist to reduce 5-FU dose,reducing its toxic side effects and provide experimental evidence.Methods: In this study,MTT assay curcumin and 5-FU against human gastric cancer MGC-803 cell line;application-based AO / EB double staining fluorescence microscopic observation and Annexin V-FITC / PI double staining flow cytometry effects of drugs on human gastric carcinoma cells MGC-803;the application based on PI staining by flow cytometry of curcumin and 5-FU against human gastric cancer MGC-803 cell cycle.Results:(1)The group combined with low dose(2.4?mol / L)5-FU and curcumin(25?mol / L)inhibits proliferation and induces apoptosis effect of MGC-803 gastric cancer cells was significantly better than the middle dose(4.8?mol / L)5-FU alone(P <0.05);(2)The group combined with dose(4.8?mol / L)5-FU and curcumin(25?mol / L)on cell proliferation andapoptosis in gastric cancer MGC-803 cells was significantly better than the high dose(9.6?mol / L)5-FU alone(P <0.05);(3)By blocking gastric cancer MGC-803 cell division cycle progression,combined with a large number of programs gastric cancer cells arrested in S phase,the group combined with curcumin and a low or middle dose of 5-FU could inhibit gastric cancer MGC-803 cell proliferation and induce apoptosis;(4)The group combined curcumin and 5-FU with anti-gastric cancer cells shows synergistic effect in a dose-dependence of time.Conclusion: This study confirms the curcumin and 5-FU on growth of cultured human gastric cancer MGC-803 cells was significantly inhibited proliferation and induced apoptosis in gastric cancer MGC-803 cells in a doseand time-dependent manner.The results for the clinical application of curcumin as 5-FU adjuvant treatment of gastric cancer,in order to reduce the dose of 5-FU,thereby reducing its toxic side effects provided a preliminary experimental basis.
