| Objective: The purpose of this study was to study the effect of xiaoyao powder+compound sanzi decoction periodic administration on the expression of anti-muuller test-tube hormone(AMH)and its type II receptor(AMHR2)in ovarian granulosa cells of PCO model rats,and to explore the mechanism of the treatment of polycystic ovary syndrome(PCOS)with kidney-tonifying and vesiculation period.Methods: 110 SPF female SD rats,random number table method to select 10 as blank control group,the remaining 100 letrozole lavage method to build model,only after the success,set up the model control group,positive control group,A medicine(xiaoyao powder),low dose group,high dose group,B medicine(compound third soup),low dose group,high dose group B drug,A medicine + B(xiaoyao powder +compound third soup)cycle use four groups,each group of 10.The positive control group was administrated for 7 days,and the other treatment groups were administrated for 12 days respectively.Six rats in each group were selected to collect right ovarian granulosa cells,and the expressions of AMH and AMHR2 in ovarian granulosa cells were detected by Western Blot and fluorescence quantitative PCR.Results: AMH protein expression: Compared with the normal group,the EXPRESSION of AMH in the granulosa cells of the PCO model group was significantly increased(P < 0.01).Compared with the model group,AMH expression in granulosa cells of ovary was significantly decreased in the low A group,high A group,low B group,high B group,high A group and positive control group(P < 0.01).The expression of AMH in granulosa cells of high B group was the lowest.Comparison between A+B periodic treatment groups: AMH expression in ovarian granulosa cells of A high group and B low group was significantly lower than that of the other three groups(P < 0.01).AMHR2 protein expression: Compared with the normal group,the EXPRESSION of AMHR2 in the granulosa cells in the PCO model group was significantly increased(P < 0.01).Compared with the model group,the expression of AMH in granulosa cells of ovary in the low A group,high A group,low B group,high B group,high A group and positive control group was significantly decreased(P < 0.01),while the expression of AMH in the high A group and low Bgroup was significantly decreased(P < 0.05).The expression of AMHR2 in granulosa cells of high B group was the lowest.Comparison between the A+B periodic treatment groups: The expression of AMHR2 in ovarian granulosa cells of the A high B low group was significantly lower than that of the other three groups(P < 0.01).AMH gene expression: Compared with the normal group,the EXPRESSION of AMH in the ovary granulosa cells of the PCO model group was significantly increased(P < 0.01).Compared with the model group,the expression of AMH in granulosa cells in all treatment groups was significantly decreased(P < 0.01).The expression of AMH gene in granulosa cells of high B group was the lowest.Compared with the other three groups,the expression of AMH in granulosa cells of the A+B group was significantly decreased(P < 0.01).AMHR2 gene expression: Compared with the normal group,the EXPRESSION of AMHR2 in the granulosa cells in the PCO model group was significantly increased(P < 0.01).Compared with the model group,the expression of AMHR2 in granulosa cells in all treatment groups was significantly decreased(P <0.01).The expression of AMHR2 in granulosa cells of high B group was the lowest.Comparison between the A+B periodic treatment groups: AMHR2 expression in ovarian granulosa cells of the A high B low group was the lowest,which was statistically different from that of the A low B low group and the A low B high group(P < 0.01).Conclusion: The expression of AMH and AMHR2 in ovarian granulosa cells of PCO model rats could be significantly reduced by the method of tonifying kidney and regulating vesicle cycle.Different concentrations and drug combinations may have different degrees of regulation on the expression of AMH and AMHR2 proteins and genes in PCO granulosa cells.The treatment of POLYcystic ovary syndrome by tonifying kidney and regulating vesicle cycle may be related to the expression of related proteins that affect follicle recruitment,development and excretion. |
