| Objective:To establish an animal model combined with the wind-phlegm blockage channel syndrome and cerebral infarction disease and verify it.To provide an animal model for the prevention and treatment of cerebral infarction with TCM.Methods:In this study,80 rats were randomly divided into the normal group(10 rats)and the model-make group(70 rats).The normal group was reared normally.The model-make group was reared high-fat diet(20g/rat/day)to replicate the sputum syndrome model,and was gavaged Fuzi decoction(2g/kg/day)to replicate the hyperactivity of liver-yang syndrome model.On the 31th day,the rats,s middle cerebral arteries were occluded with suture embolization method,so that established a model combined with the wind-phlegm blockage channel syndrome and the cerebral infarction disease.Then according to the idea of"testing the syndrome by prescription",the model was reversely verified with Zhenfangbaiwanzi decoction,and the model-maked rats were randomly divided into model control group,Chinese medicine group(gavaged by Zhenfangbaiwanzi decoction,,8.1g/kg/day),and western medicine group(injected by Edaravone,3.2mg/kg/day),17 rats in each group.After two weeks of intervention,we observed and tested the indicators of per group,including the macro symptoms and micro indicators,such as neurological deficit scores,cerebral infarction volume,blood lipid,blood rheology,NE,F,TXB2,and T3;compared the differences of various indicators,and evaluated the success of the model from various aspects and angles.Result:1.Macro symptoms:The rats in the normal group were moderate in shape and weighed between 230g and 260g.They were in good spirits,with soft and moist hair,having a normal diet,acting normally,mild temperament,no congestion in conjunctiva.The rats in the model control group were obese than the preforms and weighed between 250g and 280g.They were inferior in spirit,with dry hair,slightly increased food intake,walked on one side,moved slowly,irritable,and their degree of irritation was over Grade 2,conjunctival congestion.The rats in the traditional Chinese medicine group were moderate in shape,weighing between 220g and 280g,and their spirits were significantly improved.Their hair was moisturized,their food intake increased,their actions were more convenient,they tended to walk a bit,their actions increased in speed,their temperament was mild,and their irritability was reduced.The conjunctiva was not markedly congested.The rats in the western medicine group were moderate in shape and weighed between 230g and 270g.Their spirits improved significantly,their hair became moist,their food intake increased,their actions were more convenient,they tended to walk a bit,their speed of action increased,their temperament was still slightly irritable,and their degree of irritation was Grade1~2,conjunctiva was slightly congested.2.Neurological deficit score:Compared with the normal group,the score of the neurological deficit in the model control group was significantly higher(P<0.01).Compared with the model control group,the neurological defect scores of the Chinese medicine group and the western medicine group were significantly lower than the model control group(P<0.01),while there was no significant difference between the Chinese medicine group and the western medicine group(P>0.05).3.Brain infarction volume:Compared with the normal group,the volume of cerebral infarction in the model control group was significantly increased(P<0.01).Compared with the model control group,the volume of cerebral infarction in the traditional Chinese medicine group and western medicine group was significantly reduced(P<0.01,P<0.05).4.Hemorheology,blood lipids:(1)Hemorheology:Compared with the normal group,plasma viscosity,low-cut viscosity,middle-cut viscosity,and high-cut viscosity of the model control group were significantly increased(P<0.01).Compared with the model control group,the plasma viscosity,low-cut viscosity,middle-cut viscosity,and high-cut viscosity of the Chinese medicine group and western medicine group were significantly reduced(P<0.01).And the 4 indicators above of the Chinese medicine group were significantly lower than the western medicine group(P>0.05).(2)Blood lipids:Compared with the normal group,the levels of TC,TG,and LDL-C in the model control group increased,while the levels of HDL-C decreased(P<0.01).Compared with the model control group,the levels of TC and TG in the Chinese medicine group and the western medicine group were significantly reduced,and the HDL-C level was significantly increased(P<0.01).Compared with the traditional Chinese medicine group,the levels of TC,TG,and LDL-C in the western medicine group increased significantly,while the levels of HDL-C decreased significantly(P<0.01).5.NE,F,TXB2,T3:Compared with the normal group,F,NE,and TXB2in the model control group were significantly higher than those in the normal group,while T3was significantly decreased(P<0.01).Compared with the model control group,the levels of F,NE,and TXB2in the Chinese medicine group and the western medicine group were significantly reduced,while the T3levels were significantly increased(P<0.01).There was no significant difference between the traditional Chinese medicine group and the western medicine group(P>0.05).Conclusion:1.High fat diet,Fuzi decoction administration and blocking the middle cerebral artery with suture-occluded method can establish the animal model combined with the wind-phlegm blockage channel syndrome and the cerebral infarction disease.2.Zhenfangbaiwanzi decoction and Edaravone injection could improve the macro symptoms of model rats and the score of cerebral infarction volume nerve function defect,blood lipid and hemorheology F、NE、TXB2、T3and other microscopic indexes,and the effect was equivalent,which reversely proved that this model was the animal model combined with the wind-phlegm blockage channel syndrome and the cerebral infarction disease. |
