Purpose:To explore the effect of Chinese medicine Pidan Ning on the expression of GLUT-4 on IR and skeletal muscle cells of IGR rats.Materials and methods: 100 Specific Pathogen Free Wistar experimental rats,sex,male.Day old,between 60 and 70 days,body weight,220 ± 20 g.After 7 days of feeding,using random number method,100 rats were randomly divided into two groups: blank control group(20 rats)and model building group(80 rats).Then,the model-building rats were established as IGR rat models,and the model group rats were fed with high-calorie feed and free access to water.Rats in the blank control group were fed with conventional feed.After continuous feeding for 12 weeks,the OGTT test was performed on the rats in the model-building group.After judging that the IGR rats were successfully modeled,the rats in the successful model-building group were randomized again:model control group,Pidan Ning group and metformin control group.And intervene through the following methods: the blank control group rats and the model control group were orally gavaged with 5ml / kg of distilled water daily;The Pidan Ning group was orally gavaged with 10 g / kg Pidan Ning preparation daily;the metformin control group was orally gavaged with 150 mg / kg metformin suspension per day.The four groups intervened for 28 days.Testing the body weight,FBG and FINS of the four groups of rats before and after medication,and the IR situation was evaluated by GC technology.When we finished the experiment,the four groups of rats were sacrificed,and the expression of glucose transporter 4 in skeletal muscle cells was detected by western-blot method.Results:1.After modeling,the average value of FBG in the blank control group was 4.54 mmol / L,and the average value of 2h PG was 6.73 mmol / L.Through calculation,the maximum blood glucose FBG + 1.96 S in the blank control group was 6.5 mmol / L,and 2h PG + 1.96 S was8.69 mmol / LAfter measuring the results of 4 groups of FBG and 2h PG,the number of rats meeting the following conditions was 43:(1)7.0 mmol / L> FBG> 6.5 mmol / L or 11.1 mmol/ L> OGTT 2h PG> 8.69 mmol / L;(2)Random blood glucose <16.7 mmol / L.2.Evaluation of the IR of the model group: After modeling,the FINS concentration in the blood of rats in the model control group,spleen group and metformin control group was higher than that in the normal control group(P<0.01),and the glucose infusion rate(GIR)in the normal control group was higher than the other three groups(P <0.01),and the FINS and GIR results were comparable among the three groups(P> 0.05);after treatment with drugs,the blood FINS level in the rats of the two drug treatment groups was higher than that of the normal control group(P<0.01)but higher than the model control group To be low(P<0.01),the GIR of the two drug treatment groups was lower than that of the normal control group(P<0.01)but higher than the model control group(P<0.01).The difference between FINS and GIR between the two drug treatment groups was not significant(P> 0.05).3.Evaluation of the expression level of GLUT-4 on skeletal muscle cells of 4 groups of rats:After modeling,the expression of GLUT-4 on the skeletal muscle cells of the model control group,Pidan Ning group and metformin group was lower than that of the normal control group(P <0.01),and the GLUT-4 there was no significant difference in expression level(P> 0.05);after treatment,the expression level of GLUT-4 on skeletal muscle cells of the two drug treatment groups was lower than that of the normal control group(P <0.01),but both were higher than the model control group(P <0.01),and the difference between the two drug treatment groups was not significant(P> 0.05).Conclusion:1.A stable IGR rat model can be obtained by feeding healthy rats with high-calorie feed.2.Pidan Ning significantly improved insulin resistance in rats with impaired glucose regulation.3.Pidan Ning significantly increased the expression level of GLUT-4 on the skeletal muscle cells of rats with impaired glucose regulation.4.Pidan Ning can improve the insulin resistance of impaired glucose regulation rats by increasing the expression level of GLUT-4 on skeletal muscle cells of IGR rats.
