Purpose:To observe the effects of Euphorbia euphorbia extract combined with Chidamide on the proliferation and apoptosis of human triple-negative breast cancer cell line MDA-MB-231,and to explore the change of the inhibition rate of MDA-MB-231 cells by the combination of two drugs And changes in apoptosis and cycle effects,providing theoretical and experimental evidence for clinical applications.Material and method: 1.Research object: Experiment with log-growth triple negative breast cancer cells.2.Experimental drugs: Euphorbia euphorbia extract,Chidamide.3.Experimental method: CCK-8 method was used to determine the proliferation effect of Euphorbia euphorbia extract on breast cancer cell MDA-MB-231.The concentrations of Euphorbia euphorbia extract were 1?g / ml,5?g / ml,10?g / ml,20 ?g / ml,50 ?g / ml,75 ?g / ml,100 ?g / ml,150 ?g / ml;CCK-8 method was used to determine the proliferation effect of Chidamide on breast cancer cell MDA-MB-231,the concentration of Chidamide was respectively 0.3?M,1?M,3?M,10?M,30?M;Detect the toxic effects of Euphorbia euphorbia extract on normal human breast cells MCF-10 A,the concentrations are 1mg/ml?5mg/ml?10mg/ml?20mg/ml?50mg/ml?75mg/ml?100mg/ml?200mg/ml;To determine the effect of Euphorbia euphorbia extract combined with different concentrations of Chidamide on the proliferation of cell line MDA-MB-231,1 ?g / ml and 5 ?g / ml were used.The change of IC₅₀ was calculated by combining the extract of Euphorbia serrata with 0.3?M,1 ?M,3 ?M,10 ?M,and 30 ?M of Chidamide.Flow cytometry was used to detect the effects of IC₅₀ concentration of 1?g / ml and 5?g / ml euphorbia euphorbia extracts and Chidamide on cell cycle MDA-MB-231 cycle and apoptosis when the two drugs were combined for 48 h.Result:1.Euphorbia euphorbiae extract,Chidamide,Euphorbia euphorbiae extract and Chidamide have inhibitory effects on the cell line MDA-MB-231,and have a positive correlation with the concentration.Reached a median lethal concentration of 60.64?g/ ml in 24 hours,Reached a median lethal concentration of 30.18 ?g / ml in 48 hours;Chidamide concentration reaching a lethal dose of 24 hours at 5.69 ?M,Chidamide at a median lethal dose of 48 hours at 3.51 ?M;The effect of Euphorbia euphorbiae extract on human breast cell MCF-10 A at concentrations of 1?g / ml and 5?g / ml after 24 h and 48 h had no significant effect on the survival rate of normal breast cells.The synergistic effect of the combination of 1 ?g / ml and 5 ?g / ml extracts of euphorbia euphorbiae and Euphorbia esculenta and 0.3 ?M-30 ?M Chidamide on MDA-MB-231 cell line was detected after 48 h.Concentration reaching half lethal dose after 24 hours was 2.29 ?M.Concentration reaching half lethal dose after 48 hours was0.88?M?2.The concentration of 1 ?g / ml and 5 ?g / ml of Euphorbia euphorbiae extracts combined with 3.5 ?M of Chidamide,48 hours after MDA-MB-231 cell cycle arrest in G1 phase.3.The concentration of 1 ?g / ml and 5 ?g / ml of Euphorbia euphorbiae extract combined with Chidamide 3.5 ?M for 48 h,respectively,could enhance the apoptosis rate of MDA-MB-231 cell line induced by Chidamide.The synergy is synergistic.Conclusion:1.Compared with the single drug Chidamide,the combination of Euphorbia sibirica extract and Chidamide in human breast cancer cell line MDA-MB-231 has enhanced the inhibition of the cell line,and has a synergistic effect.2.The combination of Euphorbia lupus extract and Chidamide makes the cycle of human breast cancer cell line MDA-MB-231 blocked in G1 phase,and the extract of Euphorbia lupus can enhance the treatment of MDA-MB-231 cell blockade.3.Compared with the single drug Chidamide,the extract of Euphorbia fulvae and Chidamide can induce the apoptosis rate of human breast cancer cell line MDA-MB-231.
