Objective:To investigate whether the extract of Euphorbia lanceolata can reverse the resistance of breast cancer resistant cell line MCF-7 / ADR to adriamycin.Materials and methods:1.Research objects: Human breast cancer cell line MCF-7(luminal type B)and breast cancer adriamycin resistant cell line MCF-7 / ADR.2.Experimental drugs: Doxorubicin and Euphorbia euphorbia extract.3.Methods:(1)CCK-8 method was used to detect the cell activity of adriamycin,Euphorbia euphorbia extract and the two drugs on MCF-7 and MCF-7/ADR cells after 24 hours,and IC50 and MCF-7/ADR cells were calculated respectively Drug resistance index(RI),verify the multi-drug resistance of MCF-7/ADR cells,and calculate the low toxic dose of Euphorbia lupus(cell survival rate is more than 90%),and set it as the concentration and follow-up study Doxorubicin works together.(2)To detect the reversal effect of Euphorbia fischeriana extract on adriamycin resistance of MCF-7/ADR cells: flow cytometry was used to detect the effect of Euphorbia fischeriana extract,adriamycin alone and both drugs on the cell cycle and apoptosis of MCF-7 and MCF-7/ADR cells,and to explore the mechanism of Euphorbia fischeriana extract on adriamycin resistance of MCF-7/ADR cells.Results:1.Euphorbia euphorbia extract has inhibitory effect on MCF-7 cells and MCF-7/ADR cells,and has a dose-response relationship.IC50 is 78.52±1.87mg/m L,83.36±1.09mg/m L,There was a statistically significant difference between the two(P <0.05);when the concentration of the extract of Euphorbia vulgaris was 7.5mg/m L,the MCF-7 and MCF-7 / ADR cell survival rates were greater than 90%,which were determined as the subsequent reversed dose.2.Doxorubicin has a proliferation inhibitory effect on MCF-7 and MCF-7/ADR cells.IC50 of adriamycin on MCF-7 cells and MCF-7/ADR cells is 4.32±0.12mg/m L,68.31±0.74mg/m L,the drug resistance index was 15.81(P <0.05).3.After doxorubicin and low-dose euphorbia euphorbia extract intervened in MCF-7 and MCF-7/ADR cells,the IC50 of the two cells were 1.49±0.06mg/m L and 45.95±0.36mg/m L,respectively.The drug resistance index was 10.64,and the reversal index was 1.49(P <0.05).4.After low-dose single-drug euphorbia euphorbia extract,adriamycin,and both drugs were applied to MCF-7 cells and MCF-7/ADR cells for 24 hours,cell cycle tests were performed.Compared with the combination of the two drugs,MCF-7 cells and MCF-7/ADR cells can increase G0/G1 phase(P <0.05).The combined action of Euphorbia euphorbia extract and adriamycin can make MCF-7 cells and MCF-7/ADR cells were arrested in G0/G1 phase.5.Low-dose euphorbia euphorbiae extract and adriamycin alone and in combination were used for MCF-7 and MCF-7/ADR cells.Compared with the control group and the single drug group,the apoptosis rate in the combination group was significantly increased(P <0.05).The combined action of Euphorbia lobata extract and adriamycin can induce MCF-7 and MCF-7/ADR cells to undergo apoptosis.Conclusion:1.Euphorbia fulvae extract inhibited MCF-7,MCF-7/ADR cell proliferation,and there was a significant dose-response relationship(P <0.05).2.Low toxicity doses of Euphorbia euphorbiae extract and adriamycin can block MCF-7 cells and MCF-7/ADR cells in G0/G1 phase.3.Low toxicity doses of euphorbia euphorbiae extract and adriamycin can promote MCF-7cells and MCF-7/ADR cell apoptosis.4.The extract of Euphorbia eryngii may partially reverse the resistance of MCF-7 / ADR cells to adriamycin by inhibiting proliferation,promoting apoptosis and blocking the cells in the G0 / G1 phase.
