Purpose:Observed the influence of Electroacupuncture(EA)on the Exprenssions of Growth asso ciated protein-43,GAP-43 and Caspase-3 in rats with cerebral Ischemiaic,To explore the neurological mechanism of acupuncture treatment of cerebral ischemia-reperfusion injury,and provide new theoretical and experimental basis for clinical treatment.Material and method:select 30 healthy male SD rats,randomly divided into false surgical group(n=6),surgical group(n-24).The surgical team was modified on the basis of the classic Longa line thrombosis method to prepare the rat model of the right-hand brain arterial protoset(Middle cerebral artery occlusion,MCAO).After successful modeling(7deaths.failed5),and then preparation of ischemic re-injection model.It is divided into model groups and electro-needle groups,each with six.The pseudo-surgical group only separated the right neck total,the outer neck and the inner artery of the neck,without ligation.The electro-needle group was given a hundred-point,in-the-wall,two-sided foot threemile hole,and the model group and the fake surgical group did not intervene.After successful modeling and two weeks after the treatment,and within 24 hours before the execution,the neurological deficit of the rats was scored respectively.The immune fluorescence staining method was used to express the situation and calculate the average cell number.Results:1.There were no signs of neurological damage in the sham operation group,and the signs of neurological damage in the operation group were obvious.Compared with the sham operation group,the neurological deficit score was significantly different(P<0.01),indicating that the rats arechemic The model was successfully prepared;the neurological score of the electroacupuncture group was lower than that of the ischemic group after treatment(P<0.01)2.After modeling,the rats stayed on the rotating rod significantly less than the shamoperation group,which was statistically significant(P<0.01).Compared with the ische mic group,the rats on the electroacupuncture group stayed on the rotating rod significantly longer.There was statistical significance(P<0.05).3.Compared with the sham operation group,the expression of Caspase-3 protein in the brain tissue of the ischemic group was significantly increased(P<0.01),and the expr ession level of GAP-43 protein decreased sharply(P<0.01).The expression of Caspase-3 protein in the electroacupuncture group was slightly higher than that in thesham operation group,and significantly lower than the ischemia group(P<0.01).Compared with the ischemia group,the GAP-43 protein content in the electroacupuncture group increased significantly(P<0.05).4.Immunohistochemical results show:The number of GAP-43 positive cells in the model group dropped sharply after surgery(P<0.01),A large number of cytoplasmic brown positive signals were seen in the electroacupuncture group after treatment.Compared with the model group,the expression of GAP-43 in the electroacupuncture group was significantly increased(P<0.05).Caspase-3 positive cells were seen in the model gro up and the electroacupuncture group after surgery.All of them increased significantly,showing a dark brown color.Compared with the model group,the number of positive cells in the electroacupuncture group decreased significantly after treatment,and the difference was statistically significant(P<0.01).5.Immunofluorescence results showed that Caspase-3 positive cells with cytoplasmicblue staining and cell morphology increased explosively after modeling surgery,and decreased significantly after electroacupuncture treatment(P<0.05);A large number of green cytoplasmic positive cells were scattered in the brain tissue of healthy rats.After modeling,the number of positive cells was significantly reduced.After treatment,it returned to normal levels(P<0.01).Conclusion:1.Electroacupuncture can significantly intervene the expression of GAP-43 and Caspase-3 in the ischemic area of rats after cerebral ischemia,so as to inhibit the apoptosis of nerve cells,promote the regeneration of nerve cells.2.And the it accelerate the reconstruction of nerve function,improve the state of ischemia and hypoxia in the ischemic area.3.Electroacupuncture can improve the motor ability of rats after reperfusion.
