Objective:To observe the effect of external self-made formula combined with oral Gulong Capsule on the healing of critical radial bone defect in New Zealand rabbits,and to explore the possible mechanism of that self-made formula combined with Gulong capsule to promote the healing of bone defect.Methods: 32 healthy pure New Zealand rabbits were selected and bone defect models with 1.5cm right radius defect were made on them respectively.They were randomly divided into four groups(n = 8).Group A was applied with external self-made formula and oral Gulong capsule,Group B was externally applied with self-made formula,Group C was applied with oral Gulong capsule,and Group D was blank.The concentrations of VEGF,calcium and phosphorus in serum were detected on the day of modeling and at the end of the 2nd,4th,6th and 8th weeks after experimental intervention in each group of experimental animals.X-ray films were taken at the end of the 2nd,4th,6th and 8th weeks at the bone defect.Results:(1)At the 8th week,X-ray showed that the defect in group A was almost filled with callus,the fracture line at the broken end disappeared,and the density of new callus and the original bone bed were close and bridged with each other.The defects in group B and group C were not filled with callus,the edge of the broken end was blurred,bone density increased,and most of the broken end was bridged.In group D,the defect was not filled with callus,the broken end was slightly bridged,the bone density of new callus was lower,the broken end was slightly hardened,and some medullary cavities were closed.X-ray score: comparison between groups 6W and 8W after operation: group A,B and C scored higher than group D in each time period,with significant difference(p < 0.01);The scores of each time period in group a were higher than those in groups B and C,with significant difference(p < 0.01).The scores of group B and group C were similar in each time period,with no statistical difference(p > 0.05).Intra-group comparison: the x score of group A,group B and group C at 8W after experimental intervention was higher than that at 6W,with significant difference(p < 0.01).The x-ray scores of group D at 6W were similar to those at 8W,with no statistical difference(p > 0.05).(2)The serum concentrations of VEGF in the four groups were close before operation,and there was no statistical difference between them(P > 0.05).At the end of the 2nd week,4th week,6th week and 8th week after operation,the four groups A,B,C and D were compared,The serum concentration of VEGF in groups a,b and c was significantly higher than that in group d,p < 0.05.The serum concentration of VEGF in group a was significantly higher than that in groups b and c(p < 0.05).The serum concentration of VEGF in groups b and c was close to each other in all time periods,with no statistical difference(p > 0.05).Compared with each time point in the each group,VEGF concentration in groups A,B,C and D increased significantly at the end of 2 weeks after operation,reached the highest point at this time and then decreased gradually.At the 8th week,the VEGF concentration in group A,group B and group C was still at a higher level than that before operation,and the VEGF concentration in group D was close to that before operation at the 8th week.(3)the product of calcium and phosphorus in each group was close before operation,and there was no statistical difference between the two groups(p > 0.05).Compared with group d at 2W,4W,6W and 8W after operation,the product of calcium and phosphorus in group a,b and c was significantly higher than that in group d(p < 0.05).The product of calcium and phosphorus in group a was higher than that in groups b and c,with significant difference(p < 0.05).The product of calcium and phosphorus in groups b and c was close to each other in all time periods,with no statistical difference(p > 0.05).Conclusion:(1)External application of self-made prescription can enhance VEGF expression,prolong VEGF expression peak and promote bone healing.(2)External application of self-made prescription can promote calcium salt deposition and bone healing by increasing the product of calcium and phosphorus.(3)Oral administration of Gulong capsule can enhance the expression of VEGF,prolong the peak expression of VEGF and promote bone healing.(4)Oral administration of Gulong capsule can promote calcium salt deposition and bone healing by increasing the product of calcium and phosphorus.(5)Self-made prescription and Gulong capsule have synergistic effect.Combined use can better promote the expression of VEGF,improve the product level of calcium and phosphorus,and promote bone healing faster and better.
