Objective To compare the ability of promoting bone defectreconstruction of calcium phosphate cement(CPC) with or without fibringlue(FG) for cavitary bone defect in coccyxes of rats.Methods Took thirty-six female SD rats, which were eight weeks old,weighed about two hundred grams, divided them into two groups averagely.One group was used for building osteoporotic model (OVX group), and theother as normal bone mass (NL group). Eighteen rats of NL group wererandomly divided into experimentative subgroup (NI) and sham controlsubgroup (NN) in average. Eighteen rats of OVX group were randomly dividedinto experimentative subgroup (OI) and sham control subgroup (ON) inaverage. Used puncture needle to build cavitary bone defect(CBD) in thefirst coccyx(Co1) and the second coccyx(Co2) of four subgroups. CPCwas filled into the first coccyx (NI-Co1&OI-Co1) and CPC/FG was filledinto the second coccyx (NI-Co2&OI-Co2) of NI and OI subgroups. In the NNand ON subgroups, the first coccyx (NN-Co1&ON-Co1) and the second coccyx(NN-Co2&ON-Co2) weren’t filled any bone defect filler (BDF). Three ratswere extracted from the four subgroups each time after one week, six weeksand twelve weeks postoperation respectively. Assessed the ability ofpromoting bone defect reconstruction of CPC with or without FG for cavitary bone defect in coccyx in the two experimentative subgroups andassessed the automatic reconstructive ability in the two sham subgroupsby micro-CT and pathological analysis.Results (1) In the NL experimentative subgroup, whatever CPC/FG or CPCalone,the bone defect reconstruction of both six weeks and twelve weekspostoperation had changed significantly compared with that of one weekpostoperation(p<0.05). The bone defect reconstruction of Co2that filledwith CPC/FG were better than that of Co1at six weeks and twelve weekspostoperation (p<0.05).(2)In the OVX experimentative subgroup, whatever CPC/FG or CPC alone,the bone defect reconstruction at six weeks postoperation didn’t changesignificantly (p>0.05), but the bone defect recongstruction at twelveweeks postoperation changed significantly (p<0.05) when compared withthat of one week postoperation. The bone defect reconstruction of Co2thatfilled with CPC/FG wasn’t better than that of Co1at six weekspostoperation, but the bone defect recongstruction of Co2at twelve weekspostoperation was better (p<0.05).Conclusions (1)It’s six weeks postoperation when CPC/FG exhibitedstronger reconstructive ability than CPC alone in the NL rats, but it needs12weeks in OVX rats.(2)Whatever in the NL rats or in the OVX rats, the modified CPC,which combined with FG as macroporous maker for new bone to grow in, hasa faster resorptive rate and stronger ability of promoting bone defectreconstruction.
