| BackgroundWith the progress of human society,more and more people are exposed to ionizing radiation in their daily life,so more and more radiation damage is caused.As a reproductive system highly sensitive to ionizing radiation,male reproductive function is vulnerable to different degrees of radiation damage,such as sperm concentration,viability,vitality,morphological structure,chromosome and DNA,etc.will undergo a series of changes,which will lead to male fertility decline and progeny dysplasia.At present,there are still few studies on the protection of radiation injury by TCM compounds,while the research group’s self-designed formula for qi qi and detoxification has achieved certain efficacy in the previous studies on testicular radiation injury.Therefore,this study will continue to explore the effects of ionizing radiation on male fertility and the prevention and treatment effects of yiqi detoxification formula.ObjectiveIn terms of theoretical research,according to the traditional Chinese medicine literature,the characteristics of the theory of "kidney dominating reproduction" were summarized,and the relationship between male reproductive sperm and fetal pregnancy was discussed in combination with the theory,so as to provide theoretical guidance for the impaired fertility caused by the decline of sperm quality.In the experimental study,combined with the characteristics of ionizing radiation injury,the effects of ionizing radiation on sperm quality were observed to observe the changes in fertility of male mice and the prevention and treatment effects of yiqi detoxification formula,and to explore whether TLR5 signal played a role in the above process.MethodTheoretical research:Based on the traditional Chinese medicine theory of "kidney dominating reproduction",this paper combs the theoretical basis of "kidney dominating reproduction",then combs the relationship between kidney-procreation and male procreation,and then discusses the influence of male procreation on pregnancy,so as to provide the theoretical basis of traditional Chinese medicine for male infertility patients.Experimental Study:Experiment 1:24 male Balb/c mice were randomly divided into blank group,model group and qi-yijiedu group according to the body mass according to the random number table method.Mice in the blank group received false irradiation as the blank control,and mice in the model group and the yiqi detoxification group received one-time whole-body irradiation with 1.0gy 60Co radiation.Mice in the blank group and the model group were given deionized water 7 days before illumination and 35 days after illumination.On the 36th day,according to the ratio of male to female 1:2,each male Balb/c mouse was caged with the female Balb/c mouse for five weeks.Vaginal plugs of female Balb/c mice were checked at 8 o’clock in the morning and 5 o’clock in the afternoon every day.Pregnant female mice were isolated by weighing body mass and raised in a single cage.The male mice were sacrificed and collected on the 70th day after the shooting,and the body mass changes,testicular and epididymal coefficients and semen routine indexes of the male mice were recorded.After the birth of the offspring mice,the body mass changes of the female mice,the average litter size and the body mass changes of the offspring mice were recorded.Experiment 2:According to the body mass,72 Balb/c male mice were randomly divided into blank group,model group,andolin group and yiqi detoxification group.Mice in the blank group received false irradiation as the blank control,and mice in the model group,the andolin group and the yiqi-detoxification group received one-time whole-body irradiation with 2.0gy 60Co radiation.Mice in the blank group and model group were administrated with deionized water 7 days before illumination and 35 days after illumination,and mice in the andolin group and the yiqi-jiedu group were administrated with andolin suspension and yiqi-jiedu prescription solution respectively.On the 36th day,according to the ratio of male to female 1:2,each male Balb/c mouse was caged with the female Balb/c mouse for five weeks.Vaginal plugs of female Balb/c mice were checked at 8 o ’clock in the morning and 5 o ’clock in the afternoon every day.Pregnant female mice were isolated by weighing body mass and raised in a single cage.Male mice respectively after according to 35 d,70 d executed and materials,testing coefficient of testis and epididymis of male mice,the ultrastructural changes of testis and epididymis,complete blood count and semen routine index,after the birth of their offspring mice to observe to 42 days of age to be put to death,recording the conception rate of females,the average litter size number,sex ratio of their offspring mice,body quality,complete blood count and blood biochemical changes.Experiment 3:ATP content,sperm membrane potential changes,sperm TLR5,MyD88,IRAK4,PI3K,and GSK-3α in the sampled mice in experiment 2 were detected.ResultTheoretical research:(1)the functional basis of "kidney dominating reproduction" is based on the theory of"kidney stress essence" in the theory of TCM.Among them,"kidney stores essence" mainly reflects for the kidney has the nature of sealing property,can hide the essence of first,acquired and zang-fu essence.(2)"kidney main reproduction" is mainly reflected in two points.The first is that the kidney is closely related to male reproductive organs.The kidney can dominate male genital organs through meridians,and the kidney can be opened to the anterior Yin.The second is that the kidney can regulate the reproductive function of the human body by regulating the essence of the kidney.(3)the functions and storage sites of male reproductive sperm and renal sperm are different.Kidney essence is stored in the kidney,which can not only nourish and maintain the physiological functions of the internal organs,but also serve as the material basis for metaplastic male reproduction.But the male reproduction essence hides in the fine room,has the reproduction function,may eduction outside the body and the female reproduction essence union becomes the embryo.(4)for fetal pregnancy,the formation of fetal pregnancy is the male reproductive essence and female reproductive essence of each other,therefore,the health of male reproductive essence is a necessary factor for the formation of fetal pregnancy,its abnormality will lead to insufficient fetal endowment.Experimental Study:Experiment 1:1.0 Gy 60Co γ radiation can rapidly reduce the body mass of male Balb/c mice,and the body mass recovery can be promoted by giving mice qi supplementation and detoxification before and after irradiation.At the same time,the sperm quality of the mice and the litter size of the female mice they mated with after 70 days still did not return to the level of the blank group,and the prevention and treatment of yiqi-jiedu prescription effectively improved the above situation.Experiment 2:After a one-time whole-body irradiation of mice with 2.0 Gy 60Co γ radiation,the testicular,epididymal coefficient,sperm quality,testicular and epididymal ultrastructure,and the whole blood cell count were seriously damaged 35 days after the irradiation.After 70 days,the sperm concentration and CBC of the male mice were restored,but the sperm motility and viability were still not restored.As for the offspring mice,the litter size of the offspring mice in the model group significantly decreased,and the number of white blood cells and lymphocytes significantly increased,suggesting that the offspring mice were affected by the quality of parental sperm and developed slowly,and the prevention and treatment of yiqi-jiedu prescription effectively improved the above situation.Experiment 3:At 35 and 70 days after irradiation,the sperm membrane potential and sperm ATP content of each group of mice decreased after irradiation,and the damage of sperm membrane potential of 35 days after irradiation was effectively improved by andolin and yiqi-jiedu.The expression of TLR5,MyD88,IRAK4,PI3K and GSK-3α in mice in the 35d model group increased significantly,while the expression of TLR5 and other factors in sperm was down-regulated by the formula of yiqi-detoxification.In the testis,the expression of TLR5 in the model group was significantly lower than that in the blank group,and there was no significant difference between the administration group and the blank group.Conclusion:Theoretical research:The function basis of "kidney dominating reproduction" is based on the function of"kidney stores essence",and the kidney essence and the reproductive essence of male have the relationship of mutual root and mutual use.At the same time,fetal pregnancy is closely related to the reproductive essence of male,the reproductive essence of male filling is a necessary condition for good birth and good breeding.Thus,we can nourish the essence of reproduction by nourishing the kidney essence.Therefore,based on the basic principle of good birth and good breeding and the characteristics of kidney as the root of seal.We should give priority to prevention,attach importance to the care of the kidney,should not be presumptually excreted the kidney essence,and before the plan to have children do the necessary relevant examination,in order to ensure good embryo quality.Experimental Study:This experiment replicated the previous testicular radiation injury model of the research group,and further observed the damage of ionizing radiation on the reproductive function of male mice,and the damage of male mice’s reproductive function caused by YQJD prescription given before and after irradiation had a good prevention and treatment effect.At the same time,it was observed that ionizing radiation could cause changes in the expression of TLR5 and downstream signal factors on the sperm of mice,which may be one of the main reasons for the decrease in sperm motility caused by ionizing radiation,and the YQJD prescription to some extent regulated these signal changes.However,in combination with the testicular experiment,the same signal changes as sperm were not observed,possibly because the signaling pathway was only expressed in sperm.Therefore,the protective effect of yiqi detoxification formula on testicular radiation injury may still be achieved by activating NF-κB signal,and the specific mechanism still needs to be further explored. |
